摘  要:目的探究大肠杆菌感染时子宫内膜细胞细胞骨架的变化以及所涉及的信号通路。方法利用细菌侵袭实验分析大肠杆菌侵袭进入子宫内膜细胞的情况,以免疫荧光法观察大肠杆菌侵袭时子宫内膜细胞肌动蛋白微丝的变化,应用免疫印迹法以Akt磷酸化特异性抗体检测细胞内Akt的活化。结果大肠杆菌能够有效侵袭进入子宫内膜细胞;大肠杆菌的侵袭能够诱导子宫内膜细胞的肌动蛋白微丝发生重新组装、增强细胞内Akt磷酸化水平;Akt的抑制剂及其上游激酶PI3K的抑制剂均能维持肌动蛋白微丝的稳定并阻断大肠杆菌侵袭进入子宫内膜细胞。结论大肠杆菌通过诱导子宫内膜细胞中Akt分子发生磷酸化,引起肌动蛋白微丝发生重新组装,从而使细菌被内化至子宫内膜细胞内部。Objective To investigate the cytoskeleton changes of endometrial cells during E.coli infection and the involved signaling pathways.Methods Bacterial invasion assay was used to analyze the invasion of E.coli into endometrial cells.Immunofluorescence method was used to observe the changes of actin microfilaments in endometrial cells during the invasion of Escherichia coli,and Western blot method was used to detect the activation of Akt by Akt phosphorylation specific antibody.Results E.coli can effectively invade endometrial cells.The invasion of E.coli can induce the reassembly of actin microfilaments in endometrial cells and enhance the Akt phosphorylation level in the cells.Both Akt inhibitors and inhibitors of its upstream kinase PI3K maintain the stability of actin microfilaments and block the invasion of E.coli into endometrial cells.Conclusion E.coli induces phosphorylation of Akt molecule in endometrial cells,causing actin microfilaments to reassemble,thereby internalizing the bacteria into endometrial cells.
