摘 要:高粱中的多酚对人体健康具有积极作用,通过单因素和响应面优化超声辅助乙醇提取高粱总多酚的工艺,得到超声提取高梁多酚的最佳工艺条件为:超声时间58 min,温度57℃,超声功率450 W,料液比1∶17,在此条件下高粱多酚的得率为1.362%。通过对5种大孔树脂的静态吸附及解吸附性能比较,确定HPD-300型树脂为分离纯化高粱多酚的最佳树脂,通过对上样浓度、上样pH、上样速度、洗脱溶剂、洗脱速度的单因素实验,得到最佳纯化条件为样品质量浓度1.33 mg/mL,上样pH 4,动态吸附速度2 mL/min,乙醇洗脱剂体积分数60%,洗脱速度1.5 mL/min。经过纯化后冻干的高粱多酚粉纯度提高50.42%,是纯化前的3.3倍。通过化学定性分析、红外光谱、紫外-可见吸收光谱及液相色谱-质谱联用法检测分析,得到高粱多酚中有19种酚酸类物质,5种丹宁类物质,其中儿茶素、原儿茶酸、苯丙氨酸、咖啡酸、没食子儿茶素的含量较高。The polyphenols in sorghum have positive effects on human health.The technology of ultrasonic-assisted ethanol extraction of total polyphenols from sorghum was optimized by single factor and response surface methodology.The optimal conditions for ultrasonic extraction of sorghum polyphenols were as follows:ultrasonic time of 58 min,temperature of 57℃,ultrasonic power of 450 W,solid-liquid ratio of 1∶17.Under these conditions,the extraction rate of sorghum polyphenols was 1.362%.Through the comparison of static adsorption and desorption performance of five macroporous resins,HPD-300 resin was determined to be the best resin for separating and purifying sorghum polyphenols,and the optimal purification conditions were 1.33 mg/mL,pH 4,dynamic adsorption speed 2 mL/min,60%volume fraction of ethanol eluent,and 1.5 mL/min by single factor experiments on sample concentration,pH,elution solvent and elution speed.After purification,the purity of lyophilized sorghum polyphenol powder was increased by 50.42%,3.3 times of that before purification.Through chemical qualitative analysis,infrared spectroscopy,ultraviolet-visible absorption spectroscopy and liquid chromatography-mass spectrometry,19 phenolic acids and 5 tannins were found in sorghum polyphenols.Among them,the content of catechins,protocatechuic acid,phenylalanine,caffeic acid,and gallocatechin was higher.
