摘 要:目的探讨不同成分的饲料对葡聚糖硫酸钠(DSS)诱导溃疡性结肠炎(UC)小鼠模型的影响。方法68只小鼠随机分为2组,分别饲喂饲料A(n=42)和饲料B(n=26),适应性喂养后随机分为对照组[A_(Ctr)组(n=21)和B_(Ctr)组(n=6)]和DSS组[A_(25 DSS)组(n=21)、B_(25 DSS)组(n=6)、B_(30 DSS)组(n=6)、B_(35 DSS)组(n=8)]。各组小鼠分别饲喂相应的饲料11 d。对照组自由饮用无菌水11 d。DSS组先自由饮用相应质量浓度DSS溶液7 d,后改成饮用无菌水4 d,实验结束后评估小鼠身体质量、疾病活动度、结肠炎症状和死亡率等。结果造模结束后,A_(25 DSS)组小鼠的身体质量与B_(25 DSS)、B_(30 DSS)和B_(35 DSS)组差异无统计学意义(P>0.05)。A_(25 DSS)组的疾病活动指数显著高于B_(25 DSS)组(P<0.0001),与B_(30 DSS)组和B_(35 DSS)组差异无统计学意义(P>0.05)。A_(25 DSS)组小鼠的死亡率也显著高于B_(25 DSS)、B_(30 DSS)和B_(35 DSS)组(P<0.001)。结论营养比例高的饲料可能有利于DSS诱导的UC模型的建立。Objective To investigate the impact of different feeds on dextran sulfate sodium(DSS)-induced ulcerative colitis(UC)in mice.Methods Sixty-eight mice were randomly allocated into two groups,each receiving feeds A(n=42)and B(n=26).After adaptive feeding,they were randomly divided into control group[A_(Ctr)group(n=21)and B_(Ctr)group(n=6)]and DSS group[A_(25 DSS)group(n=21),B_(25 DSS)group(n=6),B_(30 DSS)group(n=6),B_(35 DSS)group(n=8)].Control group drank sterile water freely for 11 days.DSS group drank the corresponding concentration of DSS solution freely for 7 days,and then sterile water for 4 days.At the end of the experiment,the body mass,disease activity index(DAI),colitis symptoms and mortality were evaluated.Results After modeling,there was no significant difference in body weight among A_(25 DSS),B_(25 DSS),B_(30 DSS)and B_(35 DSS)groups(P>0.05).DAI of A_(25 DSS)group was significantly higher than that of B_(25 DSS)group(P<0.0001),but similar to B_(30 DSS)and B_(35 DSS)groups(P>0.05).The mortality was significantly higher in A_(25 DSS)group than in B_(25 DSS),B_(30 DSS)and B_(35 DSS)groups(P<0.001).Conclusion High nutrient diet may be beneficial to the establishment of DSS-induced UC model.
