热台及培养皿类型对胚胎培养液滴温度的影响

摘  要:目的比较不同类型的培养皿在不同温度热台上皿内培养液滴温度的变化情况。方法选择3种不同品牌(Vitrolife组、Oosafe组、Falcon组)的生长皿、中央孔皿、四孔皿和ICSI皿4种不同用途的培养皿作为研究材料,模拟胚胎实验室日常工作制备培养微滴,记录各类培养皿分别放在37℃和38.5℃的热台上5 min皿内培养液滴温度变化情况。结果在37℃的热台上,各种类型的培养皿内培养液温度随时间延长呈下降的趋势,与Vitrolife组相比较,Falcon组的生长皿和没有盖油的四孔皿培养液滴温度下降更快(P<0.05);与中央孔皿的Vitrolife组相比较,Oosafe组的中央孔皿内培养液温度下降更快(P<0.05);Oosafe组和Falcon组ICSI皿内培养液滴温度下降均比Vitrolife组更快(P<0.05)。而盖油后的3组四孔皿内培养液温度变化无差异(P>0.05)。在38.5℃的热台上,不盖油的四孔皿和中央孔皿内培养液温度仍然呈下降趋势,但盖油的生长皿、四孔皿和ICSI皿内培养液温度有上升的现象。与生长皿的Vitrolife组相比,Falcon组生长皿内培养液滴温度变化无差异(P>0.05),但Oosafe组生长皿温度上升相对缓慢(P<0.05);Oosafe组和Falcon组没有盖油的四孔皿和中央孔皿内培养液温度均比Vitrolife组下降更快(P<0.05);Vitrolife组的ICSI皿内培养液滴温度上升速度比另外两组更快(P<0.05)。结论不同培养皿内培养液滴达到最佳温度时所需热台设置温度可能不同。在实际工作中,工作站热台温度设定应考虑到实际使用情况和使用培养皿类型的不同,最终应以患者的胚胎发育情况和临床结局为依据。Objective:To compare the temperature changes of liquid droplets in different types of culture dishes on heating stages with different temperature.Methods:Three different brands(Vitrolife group,Oosafe group,Falcon group)of growth dishes,center-well dishes,four-well dishes and ICSI dishes were selected as the study objects.The different types of culture dishes were prepared to replicate the procedures performed by embryologists.The temperature changes of culture droplets were recorded within 5 minutes on the heating stage at 37℃and 38.5℃respectively.Results:On the heating stage at 37℃,the temperature of the culture medium in various types of culture dishes was gradually decreasing with time.Compared with the Vitrolife group,the culture medium temperature decreased more rapidly in growing dish and four-well dish without cover oil in Falcon group(P<0.05).The temperature of the culture medium in the centre-well dish decreased faster in the Oosafe group than that in the Vitrolife group(P<0.05).The culture droplet temperature of ICSI dishes in Falcon and Oosafe groups dropped faster than that in Vitrolife group(P<0.05).However,there was no significant difference in the culture droplet temperature of three groups of four-well plates after oil(P>0.05).Additionally,on the heating stage at 38.5℃,the temperature of the culture medium in the four-well dish without the oil and the central well dish continued to decrease,while the temperature in the oil-covered growth dish,four-well dish and ICSI dish increased.There was no significant difference in the culture droplet temperature of growth dish between the Falcon group and Vitrolife group(P>0.05),while the temperature of growth dish in the Oosafe group increased relatively slowly(P<0.05).Furthermore,both Falcon and Oosafe group exhibited a more rapid loss of the medium temperature of four-well dish without the oil and centre-well dish compared to Vitrolife group(P<0.05).The temperature of the culture droplet in the ICSI dish in the Vitrolife group increased faster than
