摘 要:目的:通过网络药理学探究黄芪注射液(AI)治疗脊髓损伤(SCI)的潜在靶点和作用机制,为SCI的治疗及新药研发提供参考依据。方法:检索文献获得AI的药物活性成分,通过PharmMapper数据库获得药物活性成分靶点,结合GeneCards数据库获得SCI的潜在靶点,通过韦恩图(Venny)得到药物与疾病的交集靶点,将交集靶点上传至STRING数据库对其进行蛋白质-蛋白质相互作用(PPI)网络分析构建“药物-成分-靶点-疾病”网络图,对交集靶点进行基因本体(GO)功能和京都基因与基因组百科全书(KEGG)通路富集分析。结果:经筛选后获得289个AI靶点、3 332个SCI靶点及157个药物-疾病共同靶点,AI治疗SCI的关键靶点为白蛋白(ALB)、丝氨酸/苏氨酸蛋白激酶(AKT1)、表皮生长因子受体(EGFR)、非受体酪氨酸激酶(SRC)、胱天蛋白酶3(CASP3)。GO功能和KEGG通路富集分析显示,AI主要参与对激素的反应、细胞迁移的正调控、生殖结构发育、对细菌的反应等生物过程影响SCI,以及通过癌症途径、前列腺癌途径、脂质与动脉粥样硬化途径、粘合连接途径、膀胱癌途径等信号通路调节SCI。结论:AI可通过多靶点、多通路抑制SCI的发生和发展。Objective:To investigate the potential role of Huangqi injection in the treatment of spinal cord injury(SCI),which may provide a reference for the treatment of SCI and new drug development.Methods:The active ingredients of Astragalus were collected from the published literature.Potential target genes of Huangqi and SCI were retrieved from PharmMapper and GeneCards databases,respectively.After crossing the target genes of Astragalus and SCI through Venn diagram,the intersectional target genes were uploaded to STRING database for the protein-protein interaction network analysis,followed by a construction of drug-active component-target-disease network.Gene ontology(GO)functional enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis of intersectional target genes were performed using Metascape database.Results:There were 289 Astragalus targets,3332 SCI targets and 157 drug-disease co-targets were obtained for the action of Astragalus on SCI,including albumin(ALB),AKT serine/threonine kinase 1(AKT1),epidermal growth factor receptor(EGFR),non-receptor tyrosine kinase(SRC),and cysteinyl aspartate specific proteinase 3(CASP3).Go analysis mainly involved the response to hormone,positive regulation of cell migration,reproductive structure development,response to bacterium.KEGG pathways mainly involved the pathways in cancer,prostate cancer,lipid and atherosderosis,adherens junction,bladder cancer.Conclusion:Astragalus may suppress the onset and progression of SCI through multiple targets and pathways.
