归肾丸调控TGF-β/Snail信号通路介导上皮间质转化抑制宫腔粘连的研究

摘  要:目的研究归肾丸对原代子宫内膜上皮细胞(Primary endometrial epithelial cells,PEEC)纤维化及对TGF-β/Snail信号通路介导上皮间质转化(Epithelial mesenchymal transition,EMT)的调控作用,以阐明其对宫腔粘连(Intrauterine adhesions,IUA)的潜在治疗作用及分子机制。方法利用TGF-β1体外诱导PEEC 48 h以发生EMT,再使用归肾丸含药血清处理PEEC 24 h,通过定量PCR(Quantitative polymerase chain reaction,qPCR)和蛋白质印迹(Western Blot,WB)分别检测TGF-β1下游基因Snail,EMT标志物Vimentin、E-cadherin、N-cadherin,细胞纤维化标志物α-SMA的mRNA水平和蛋白水平的表达。结果归肾丸含药血清经验证无细胞毒性。TGF-β1成功诱导了PEEC发生EMT,表现为Snail、Vimentin、N-cadherin和α-SMA的表达升高,及E-cadherin的表达降低。归肾丸血清抑制了PEEC的EMT并降低了PEEC的纤维化水平,表现为Snail、Vimentin、N-cadherin和α-SMA的表达降低,及E-cadherin的表达升高。结论归肾丸可通过抑制TGF-β/Snail信号通路介导的EMT降低PEEC纤维化水平以治疗宫腔粘连。Objective To study the regulatory effects of Guishen Pill on fibrosis of primary endometrial epithelial cells(PEEC)and epithelial-mesenchymal transition(EMT)mediated by TGF-β/Snail signaling pathway and to elucidate the potential therapeutic effect of Guishen Pill on intrauterine adhesions and its molecular mechanism.Methods TGFβ1 was used to induce EMT of PEEC in vitro for 48 hours.Then the PEEC were treated with Guishen Pill containing serum for 24 hours.The mRNA and protein expression of TGF-β1 downstream gene Snail,EMT markers vimentin,E-cadherin,and N-cadherin,and cell fibrosis markerα-SMA were detected by quantitative polymerase chain reaction(qPCR)and Western blot(WB),respectively to characterize the therapeutic effect of Guishen Pill.Results The serum containing Guishen Pill was verified to have no cytotoxicity.TGF-β1 successfully induced the occurrence of EMT in PEEC,which was manifested as expression increases of Snail,vimentin,N-cadherin,andα-SMA and expression decrease of E-cadherin.Guishen Pill containing serum inhibited the EMT in PEEC and reduced the fibrosis level of PEEC,which was reflected by expression decreases of Snail,vimentin,N-cadherin,andα-SMA and expression increase of E-cadherin.Conclusion Guishen Pill showed therapeutic effect on intrauterine adhesions by reducing fibrosis level of PEEC via inhibiting EMT mediated by TGF-β/Snail signaling pathway.
