摘 要:目的:了解Nox抑制剂APX-115 free base对H9c2细胞抵抗高浓度葡萄糖的保护效果。方法:通过CCK8法检测H9c2细胞在不同浓度APX-115 free base预处理后的增殖活力;Western blot法和免疫荧光法检测Nox4蛋白的表达;并观察细胞形态的变化。结果:与Model组相比,APX-115 free base浓度为8μmol/L时,细胞增殖活力下降,其他浓度时,细胞增殖活力上升;APX-115 free base各处理组的Nox4蛋白表达均下调;APX-115 free base 5μmol/L处理组的细胞形态恢复梭形,细胞分布逐渐均匀,细胞间空隙减少,细胞数量基本恢复到正常水平。结论:5μmol/L时的APX-115 free base对高糖诱导下H9c2细胞的保护效果最佳。Objective:To investigate the protective effect of Nox inhibitor APX-115 free base on H9c2 cells against high concentration of glucose.Methods:The proliferative activity of H9c2 cells after pretreatment with different concentrations of APX-115 free base was detected by CCK8 assay.The expression of Nox4 protein was detected by Western blot and immunofluorescence.The morphological changes of H9c2 cells were observed.Results:Compared with the Model group,whrn the concentration of APX-115 free base was 8μmol/L,the cell proliferation activity decreased,and the cell proliferation activity increased at other concentrations.The expression of Nox4 protein in each treatment group of APX-115 free base was down-regulated.The cell morphology of the APX-115 free base 5μmol/L treatment group returned to spindle shape,the cell distribution was gradually uniform,the intercellular space was reduced,and the number of cells basically returned to normal level.Conclusion:When APX-115 free base is 5μmol/L,it has the best protective effect on H9c2 cells induced by high glucose.
