摘 要:少孢节丛孢菌(Arthrobotrys oligospora,AO)是一种重要的捕食线虫性真菌,具有防控动物线虫病的潜在应用前景。为了探究AO侵染线虫的分子机制,拟对几丁质诱导的AO和未诱导的AO菌丝进行mRNA转录组学测序和验证,利用GO和KEGG富集方法分析探究AO差异表达基因(DEGs)及其参与的代谢过程和关键信号通路,分析差异表达基因在AO响应几丁质诱导mRNA表达谱特征。结果显示,AO响应几丁质诱导后,共有2904个DEGs,其中1376个上调,1528个下调;RT-qPCR结果显示,挑选的6个DEGs表达变化趋势与测序结果一致;GO分析发现,细胞代谢过程、碳水化合物代谢过程、氮化合物代谢过程等差异明显;KEGG分析显示,DEGs主要参与真核生物核糖体的生物发生、果糖和甘露糖代谢、几丁质酶基因、C源和N源吸收途径、信号传导、细胞壁合成及捕食器官形成等信号通路。该研究探究AO响应几丁质诱导的mRNA表达谱特征分析,为深入揭示少孢节丛孢侵染线虫的分子机制奠定前期研究基础。Arthrobotrys oligospora(AO)is an important nematode-trapping fungus with potential application in the prevention and control of animal nematodiasis.To explore the molecular mechanism of AO infection in C.elegans,mRNA transcriptomics sequencing and verification were performed on chitin-induced AO and uninduced AO hyphae.GO and KEGG enrichment analyses were used to explore AO differentially expressed genes(DEGs)and their associated metabolic processes and signaling pathways in response to chitin induction.This study analyzed the characteristics of AO’s differential gene expression profile in response to chitin induction.The results showed 2904 DEGs in AO responding to chitin induction,with 1376 up-regulated and 1528 down-regulated genes.The results of RT-qPCR showed that the expression patterns of six DEGs were consistent with the sequencing results.GO analysis showed that there were significant differences in cell metabolic process,carbohy drate metabolic process and nitrogen metabolic process.KEGG analysis showed that DEGs were mainly involved in eukaryotic ribosome biogenesis,fructose and mannose metabolism,chitinase genes,C source and N source absorption pathways,signal transduction,cell wall synthesis and predatory organ formation.This study elucidated the characteristics of AO response to chitin-induced mRNA expression profile,and laid a foundation for further investigations into the molecular mechanism of Arthrobotrys oligospora infection in nematodes.
