摘 要:目的 制备一种可重复利用型磁性纳米粒子Fe_(3)O_(4)固定化纤维素酶提取灵芝中多糖和三萜的成分,提高灵芝多糖和三萜的提取率及纤维素酶的重复利用率。方法 通过制备磁性纳米Fe_(3)O_(4),合成磁性纳米Fe_(3)O_(4)@SiO_(2),用APTES进行功能化修饰,得到氨基修饰的纳米粒子,两步法固定化纤维素酶。测量固定化酶的相对酶活性进行纤维素酶的重复利用性实验,采用酶辅助超声先水提再醇提提取灵芝有效成分。采用紫外分光光度计通过蒽酮-硫酸测定灵芝多糖含量、齐墩果酸测定灵芝三萜含量。结果 固定化酶相对活性循环5次相对酶活性在100%左右,灵芝提取物多糖提取率为5.3%,三萜提取率为2.0%。结论 磁性Fe_(3)O_(4)纳米粒子载体固定化纤维素酶具有良好的可重复利用性,减少纤维素酶的使用量。使用超声辅助固定化酶经过水提加乙醇提取有效提高了灵芝多糖及三萜的提取率。OBJECTIVE To prepare a reusable magnetic nanoparticle Fe_3O_(4) immobilized cellulase to extract polysaccharide and triterpene components in ganoderma lucidum,improve the extraction rate of polysaccharide and triterpenes and the reuse rate of cellulase.METHODS By preparing magnetic nano Fe_3O_4,synthesized magnetic nano Fe_3O_4@SiO_2,and functionalized with APTES,we immobilized cellulase.The relative enzyme activity of the immobilized enzyme was measured for the reutilization experiment of cellulase,and the effective ingredients of Ganoderma lucidum were extracted by enzyme-assisted ultrasound.The content of Ganoderma lucidum polysaccharide and onolic acid were determined by anthracenone-sulfuric acid using a UV spectrophotometer.RESULTS The relative enzyme activity was about 100%,the polysaccharide extraction rate of Ganoderma lucidum extract was 5.3% and triterpenes was 2.0%CONCLUSION Magnetic Fe_3O_(4) nanoparticle carrier immobilized cellulase has good reusable properties and reduces the amount of cellulase used.The use of ultrasound-assisted immobilization enzyme has effectively improved the extraction rate of ganoderma polysaccharide and triterpenes.
