摘 要:目的探究溶质载体家族16成员11(SLC16A11)基因单核苷酸多态性与妊娠糖尿病(GDM)易感性的关系,并进一步探索SLC16A11与孕前体重指数(BMI)交互作用对GDM的影响。方法以2022年1月至2023年6月在天津市西青医院参加产前检查的1289名孕妇为研究对象,检测SLC16A11基因rs13342232位点基因多态性。Logistic回归分析SLC16A11基因多态性与GDM易感性之间的关系,交互作用分析不同基因型与孕前BMI联合作用对GDM患病风险的影响。结果1289名孕妇中有260人被诊断为GDM,其余1029人为非GDM孕妇。rs13342232位点多态性在共显性模型(AG比AA,OR=1.51,95%CI:1.10~2.06,P=0.010)、显性模型(OR=1.48,95%CI:1.09~2.02,P=0.013)、超显性模型(OR=1.51,95%CI:1.10~1.85,P=0.011)和加性模型(OR=1.40,95%CI:1.05~1.86,P=0.021)中均与GDM的患病风险显著相关。rs13342232基因型与孕前BMI存在相加交互作用(AP 0.40,95%CI:0.05~0.74),AG/GG基因且孕前BMI大于24 kg/m2的孕妇罹患GDM的风险更高(OR=2.27,95%CI:1.44~3.54,P<0.001)。结论SLC16A11基因rs13342232位点单核苷酸多态性与GDM遗传易感性存在显著关联。rs13342232基因型与孕前BMI之间存在相加交互作用,AG/GG基因型且孕前超重的孕妇罹患GDM的风险更高。ObjectiveTo investigate the relationship between single nucleotide polymorphisms of solute carrier family 16 member 11(SLC16A11)gene and susceptibility to gestational diabetes mellitus(GDM).The interaction between SLC16A11 and pre-pregnancy BMI on GDM was further explored.MethodsA total of 1289 pregnant women who participated in GDM screening in Xiqing District from January 2022 to June 2023 were recruited.SLC16A11 gene rs13342232 was genotyped.Logistic regression was used to analyze the relationship between SLC16A11 gene polymorphism and GDM susceptibility,and interaction analysis was used to explore the combined effect of different genotypes and pre-pregnancy BMI on the risk of GDM.ResultsGDM was diagnosed in 260 of 1289 pregnant women.The polymorphism of rs13342232 locus was significantly associated with co-dominant model(AG vs.AA,OR=1.51,95%CI:1.10-2.06,P=0.010),dominant model(OR=1.48,95%CI:1.09-2.02,P=0.013),overdominant model(OR=1.51,95%CI:1.10-1.85,P=0.011)and additive model(OR=1.40,95%CI:1.05-1.86,P=0.021)were significantly associated with the risk of GDM.Additive interaction was found between rs13342232 genotype and pre-pregnancy BMI(AP 0.40,95%CI:0.05-0.74).Pregnant women with AG/GG genotype and pre-pregnancy BMI greater than 24 kg/m 2 had a higher risk of GDM(OR=2.27,95%CI:1.44-3.54,P<0.001).ConclusionSLC16A11 gene rs13342232 polymorphism is significantly associated with the genetic susceptibility to GDM.There is additive interaction between pre-pregnancy BMI and rs13342232 genotype.Pregnant women with AG/GG genotype and pre-pregnancy overweight have a higher risk of GDM.
