摘 要:目的采用孟德尔随机化(MR)的方法研究慢性肾脏病(CKD)和化脓性汗腺炎(HS)的双向因果关系。方法对全基因组关联性研究的汇总数据进行分析,CKD数据包括41395例患者和439303例对照组,HS数据包含409例患者和211139例对照组。本研究采用逆方差加权法(IVW)作为主要分析方法,使用Cochran Q检验评估异质性,并使用MR-Egger截距法和MR-PRESSO全局检验评估水平多效性。结果IVW固定效应模型预测CKD与HS之间呈正向因果效应(OR=2.04,95%CI:1.15~3.61,P=0.01<0.05),肾小球滤过率和HS的因果关系不显著;反向MR分析的结果不支持反向因果关系。结论CKD可能会增加HS的发病风险,而eGFR的变化不影响HS,HS对CKD不存在显著的因果关系。Objective To investigate the bidirectional causality between chronic kidney disease(CKD)and hidradenitis suppurativa(HS)using the Mendelian randomization(MR)approach.Methods The aggregate data of the whole gene-wide association study were analyzed,the data for CKD included 41395 patients and 439303 controls.The data for HS included 409 patients and 211139 controls.The inverse variance weighting(IVW)method was used as the main method of analysis in this study.The heterogeneity was evaluated using the Cochran Q test,and the horizontal pleiotropy was assessed by the MR-Egger intercept method and MR-PRESSO global test.Results The IVW method fixed-effects model predicted a positive causal effect between CKD and HS(OR=2.04,95 CI:1.15-3.61,P=0.01<0.05).There was no convincing evidence of causality between glomerular filtration rate and HS.The results of reverse MR analysis didn′t support reverse causation.Conclusion CKD may increase the risk of HS,whereas changes in eGFR do not affect HS.And there was no significant causal relationship between HS and CKD.
