摘 要:为探究促性腺激素抑制激素(GnIH)对鸡卵巢颗粒细胞葡萄糖摄入和糖酵解的影响,本试验使用不同浓度(0、10^(-10)、10^(-8)和10^(-6)mol/L)GnIH处理体外分离培养的鸡卵巢颗粒细胞,12 h后分别利用葡萄糖(GLU)试剂盒和丙酮酸测定试剂盒对细胞内葡萄糖和丙酮酸含量进行检测,并通过实时荧光定量聚合酶链式反应(qPCR)检测葡萄糖转运蛋白(GLUT1和GLUT3)以及糖酵解关键限速酶己糖激酶(HK)、磷酸果糖激酶(PFK)和丙酮酸激酶(PK)基因mRNA表达量。结果显示,与GnIH未处理组相比,10^(-8)mol/L GnIH处理组颗粒细胞葡萄糖和丙酮酸含量均显著增加(P<0.05),GLUT1和GLUT3基因mRNA表达量均极显著增加(P<0.001或P<0.01),HK、PFK和PK基因mRNA表达量均显著增加(P<0.05、P<0.001或P<0.01);10^(-6)mol/L GnIH处理组葡萄糖含量显著增加(P<0.05),丙酮酸含量显著减少(P<0.05),GLUT3基因mRNA表达量显著增加(P<0.05),HK和PFK基因mRNA表达量均显著增加(P<0.05或P<0.01)。结果表明,GnIH通过上调鸡卵巢颗粒细胞中葡萄糖转运蛋白GLUT1和GLUT3以及糖酵解关键限速酶HK、PFK和PK基因mRNA表达量,从而影响颗粒细胞对葡萄糖的摄入和糖酵解活性,最终导致颗粒细胞糖代谢紊乱。本试验可为阐明卵泡发生能量需求机制和雌性动物生殖功能障碍提供理论依据。To investigate the effects of gonadotropin-inhibitory hormone(GnIH)on glucose uptake and glycolysis in chicken ovarian granulosa cells,this study treated isolated and cultured chicken ovarian granulosa cells with different concentrations of GnIH(0,10^(-10),10^(-8),and 10^(-6) mol/L).After 12 hours,glucose and pyruvate contents in the cells were detected using glucose(GLU)and pyruvate assay kits.Additionally,the mRNA expression levels of glucose transporters(GLUT1 and GLUT3)and key rate-limiting glycolytic enzymes,including hexokinase(HK),phosphofructokinase(PFK),and pyruvate kinase(PK),were measured using real-time quantitative polymerase chain reaction(qPCR).The results showed that,compared with the GnIH untreated group,the group treated with 10^(-8) mol/L GnIH exhibited significantly increased glucose and pyruvate contents(P<0.05),with highly significant increases in the mRNA expression levels of GLUT 1 and GLUT 3(P<0.001 or P<0.01),as well as significant increases in HK,PFK,and PK gene expression(P<0.05,P<0.001,or P<0.01).In the 10^(-6) mol/L GnIH treatment group,glucose content was significantly increased(P<0.05),pyruvate content was significantly reduced(P<0.05),GLUT 3 mRNA expression was significantly increased(P<0.05),and HK and PFK mRNA expression levels were significantly increased(P<0.05 or P<0.01).These results indicate that GnIH affects glucose uptake and glycolytic activity in granulosa cells by upregulating the mRNA expression of GLUT 1,GLUT 3,and the glycolytic rate-limiting enzymes HK,PFK,and PK in chicken ovarian granulosa cells,ultimately leading to disordered glucose metabolism in granulosa cells.This study provides a theoretical basis for understanding the energy requirements in folliculogenesis and reproductive dysfunction in female animals.
