摘 要:为了明确湖北省某鸡场疑似腺胃炎病鸡发病原因,本试验对病鸡进行解剖,观察各组织病变情况;对腺胃、肝脏、法氏囊、胸腺等组织进行组织病理学检查;对腺胃石蜡切片进行禽白血病病毒(ALV)-p27抗原的免疫组织化学染色;对腺胃匀浆进行病原检测和ALV-p27抗原检测;将过滤除菌后的腺胃匀浆接种于鸡胚成纤维细胞(DF-1)分离病毒,并进行ALV亚群鉴定。结果显示,剖检可见病鸡的腺胃肿大,腺胃壁增厚,质软,腺胃乳头扁平消失;腺胃黏膜下层不同程度的淋巴细胞浸润,腺小管上皮增生,腺上皮细胞脱落坏死;其他器官不同程度的变性、坏死和炎性细胞浸润。腺管上皮胞质内出现p27抗原阳性信号。腺胃匀浆中扩增出ALV的特异性条带,同时酶联免疫吸附试验(ELISA)检测为p27抗原阳性。DF-1细胞培养物的ALV亚群鉴定为A亚群(ALV-A)和J亚群(ALV-J)共感染。结果表明,该鸡场鸡只是由ALV-A和ALV-J共同引起的腺胃炎。To investigate the cause of suspected proventriculitis in a chicken farm in Hubei Province,China,we conducted necropsies on diseased chickens and examined pathological changes in various tissues.Histopathological analysis was performed on tissues including the proventriculus,liver,bursa of Fabricius,and thymus.Immunohistochemical staining for avian leukosis virus(ALV)p27 antigen was conducted on paraffin sections of the proventriculus,and pathogenic and p27 antigen detection assays were performed on proventricular homogenates.Filtered proventricular homogenates were also inoculated into chicken embryo fibroblast(DF-1)cells for viral isolation and ALV subgroup identification.The results showed enlarged proventriculi in the diseased chickens,with thickened,softened walls and flattened,absent proventricular papillae.Submucosal lymphocytic infiltration,proliferation of glandular epithelial cells,and necrosis of glandular epithelium were observed in the proventriculus,along with varying degrees of degeneration,necrosis,and inflammatory infiltration in other organs.Positive p27 antigen signals were detected in the cytoplasm of the glandular epithelium,with ALV-specific bands amplified from the proventricular homogenates and p27 antigen positivity confirmed by enzyme-linked immunosorbent assay(ELISA).ALV subgroup identification in DF-1 cell cultures revealed co-infection with ALV subgroup A(ALV-A)and subgroup J(ALV-J).These results indicate that proventriculitis in this farm's chickens was caused by co-infection with ALV-A and ALV-J.
