一株猫源奇异变形杆菌的分离鉴定及耐药性分析

摘　　要：为探究猫腹泻的发病原因,该试验从腹泻猫的肛拭子中分离到1株细菌,通过革兰氏染色镜检、迁徙生长试验、16S rRNA PCR扩增和序列分析对分离菌株进行鉴定,并进行了溶血性试验、药敏试验、耐药基因检测及动物致病性试验。结果表明:分离菌为革兰氏阴性短杆菌,在LB琼脂和血琼脂培养基上均呈现典型的迁徙生长现象,与奇异变形杆菌的形态、生长特征相符。该菌与GenBank中犬源奇异变形杆菌(MW767054.1)的同源性为99.93%,表明该分离菌株为奇异变形杆菌。分离菌无溶血活性,其对头孢曲松、头孢氨苄、恩诺沙星、环丙沙星、阿奇霉素、磺胺甲唑、庆大霉素、多西环素及甲硝唑耐药,共检出CTX-M、aac C4、aac(6′)-Ⅰb-cr、sulⅠ和sulⅡ5种耐药基因。腹腔注射浓度为1×10^(9)CFU/mL的分离菌菌液0.3 mL可致小鼠发病并死亡。该实验成功从腹泻猫中分离到一株奇异变形杆菌,该菌对小鼠具有致病性并呈现广谱耐药。In order to explore the cause of diarrhea in cats,a strain of bacterium was isolated from the anal swab sample of a cat with diarrhea.The isolated bacteria was identified by Gram staining,microscopic examination,motility growth test,16S rRNA gene amplification and sequencing.Hemolytic activity test,drug sensitivity test drug resistance gene detection and animal pathogenicity test were performed.The results showed that the isolated strain were Gram-negative short rods and displayed typical threaded migratory growth on both LB agar and blood agar media,consistent with the morphology and growth characteristics of Proteus mirabilis.The homology of the isolate with Proteus micrabilis(MW767054.1)from dogs in Genbank was 99.93%,which was finally confirmed as Proteus mirabilis.The isolated strain was not hemolytic,resistant to ceftraxone,cephalexin,enrofloxacin,ciprofloxacin,azithromycin,sulfamethoxazole,gentamicin,doxycycline and metronidazole.Five drug resistance genes CTX-M,aac C4,aac(6′)-Ⅰb-cr,sulⅠand sulⅡwere detected in the isolated strain.Intraperitoneal injection of the isolated bacteria with 1×10^(9)CFU/mL caused morbidity and death in mice.Astrain of Proteus mirabilis was successfully isolated from the diarrhea cats,which was pathogenic and showed widesprad drug resistance.
