摘　　要：目的建立反相高效液相色谱(RP-HPLC)同时检测金花明目丸中五味子醇甲、芍药苷和京尼平苷酸含量的分析方法。方法色谱柱:Hypersil Hypurity ODS C_(18)色谱柱(250 mm×4.6 mm,5μm);柱温:30℃;流动相A:乙腈;流动相B:0.3%磷酸溶液;梯度洗脱,流速:1.0 ml/min;检测波长:五味子醇甲为249 nm,芍药苷为230 nm,京尼平苷酸为254 nm;进样体积:10μl。结果五味子醇甲、芍药苷和京尼平苷酸分别在0.2991~29.91,0.05802~5.802,0.0981~9.81μg/ml浓度范围内具有良好的线性,平均回收率分别为100.18%,100.15%和100.42%;精密度和重复性试验的RSD均在5.0%以内。结论此方法前处理简单,且相较于2020年版《中国药典》方法,专属性和重复性更佳,对于提高国家标准具有参考价值。Objective To establish a reversed phase high performance liquid chromatography(RP-HPLC)method for the simultaneous determination of schisandryl methyl,paeoniflorin and genipionic acid in Jinhua Mingmu Pills.Methods Hypersil Hypurity ODS C_(18) column(250 mm×4.6 mm,5μm)was used,and the column temperature was 30℃.The mobile phase for gradient elution was consisted of acetonitrile(A)-0.3%phosphoric acid solution(B),and the flow rate was 1.0 ml/min.The detection wavelengths were set at 249 nm for schisandryl methyl,230 nm for paeoniflorin,and 254 nm for genipionic acid,respectively.The injection volume was 10μl.Results Schisandryl methyl,paeoniflorin and genipionic acid showed good linear relationships in the concentration range of 0.2991-29.91,0.05802-5.802,0.0981-9.81μg/ml,respectively.The average recoveries were 100.18%,100.15%and 100.42%,respectively.The RSDs of precision tests and repeatability tests were both less than 5.0%.Conclusion This method not only has the advantages of simple pre-processing,but also has better specificity and repeatability compared with the methods listed in Chinese Pharmacopoeia(2020 Edition),which has reference value for improving national standards.
