苦参及其8种不同炮制品的体外抗炎、对金黄色葡萄球菌和白色念珠菌的抑制作用比较研究

摘  要:比较苦参及其8种不同炮制品的体外抗炎、抑菌作用,为苦参不同炮制品在饲料中的添加使用提供依据。采用脂多糖(LPS)刺激RAW_(264.7)小鼠巨噬细胞为体外抗炎模型,应用CCK-8法测定细胞增殖率,ELISA法检测各组对LPS刺激巨噬细胞后的肿瘤坏死因子(TNF-α)、白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)及白细胞介素-10(IL-10)的含量;采用96孔板微量稀释法测定药物对金黄色葡萄球菌和白色念珠菌的最小抑菌浓度(MIC),采用纸片扩散法测定抑菌圈直径大小。结果显示:苦参及其不同炮制品均能降低LPS刺激后细胞中TNF-α、IL-1β、IL-6、IL-10的含量,麸炒苦参和米泔水苦参较生苦参指标极显著降低(P<0.01);苦参及其不同炮制品对两种菌均有体外抑制作用,麸炒苦参、苦参炭和米泔水苦参的MIC小于生苦参,对白色念珠菌的抑菌圈显著大于生苦参(P<0.05);麸炒苦参、米泔水苦参对金黄色葡萄球菌的抑菌圈显著大于生苦参(P<0.05)。苦参及其不同炮制品均具有较好的体外抗炎及抑菌作用,其中麸炒苦参及米泔水苦参优于生苦参,因此,在动物饲料中可以添加麸炒苦参或米泔水苦参,为后续苦参炮制品在饲料领域的应用提供思路。Compare the anti-inflammatory and antibacterial effects of Sophora flavescens and its eight different processed products,providing experimental basis for the use of different processed products of So⁃phora flavescens in feed additives.Using lipopolysaccharide(LPS)to stimulate RAW_(264.7) mouse macrophages as an in vitro anti-inflammatory model,the proliferation rate of cells was determined by CCK-8 method.The contents of TNF-α,IL-1β,IL-6 and IL-10 were determined by ELISA.The contents of TNF-α,IL-1β,IL-6 and IL-10 in LPS stimulated cells were reduced by Sophora flavescens and different processed products.Bran-processed and rice-washed processed are significantly lower than raw products(P<0.01).Sophora flavescens and its different processed products have inhibitory effect on both bacteria,the MIC of carbonized-processed,branprocessed and rice-washed processed was less than that of raw Sophora flavescens,and the bacteriostasis circle was larger than that of raw Sophora flavescens(P<0.05).Sophora flavescens and its different processed products all have good antiinflammatory and antibacterial effects,among which bran-processed and rice-washed processed are better than raw Sophora flavescens.Therefore,bran-processed and rice-washed processed can be added to animal feed,providing ideas for the subsequent application of Sophora flavescens processed products in the feed field.
