扶芳藤中总黄酮提取工艺的优化及抑菌活性研究

摘  要:研究旨在探讨扶芳藤中总黄酮的提取工艺及抑菌效果,为扶芳藤作为中草药饲料添加剂应用提供参考依据。采用正交试验结合紫外分光光度法,以扶芳藤总黄酮含量为评价指标,考察乙醇浓度、料液比、提取时间、提取次数4个因素对扶芳藤总黄酮提取率的影响,优选最佳提取工艺,并采用滤纸片浸渍法评价抑菌活性。结果显示:扶芳藤总黄酮浓度为0.015~0.04 mg/mL时线性关系良好(R^(2)=0.9994),平均加样回收率为99.83%(RSD<2%),精密度、稳定性良好;扶芳藤总黄酮最佳提取工艺为用70%乙醇作提取剂,料液比1∶40,60℃超声提取2次,每次90 min,提取率为17.85%。扶芳藤总黄酮对革兰氏菌有明显的抑制作用,在5~25 mg/mL范围内,随浓度增加抑菌作用增强,在相同浓度下对不同菌株的抑制作用大小为大肠杆菌>金黄色葡萄球菌>痢疾杆菌。优化的提取工艺绿色便捷,稳定可靠,提取得到的总黄酮对大肠杆菌、金黄色葡萄球菌、痢疾杆菌均有明显的抑制作用。The research is dedicated to exploring the extraction process and antibacterial efficacy of total flavonoids in Euonymus fortunei,furnishing a reference foundation for the application of Euonymus fortu⁃nei as a Chinese herbal feed additive.Through orthogonal experiments coupled with ultraviolet spectrophotometry,taking the content of total flavonoids as the evaluation index,the influences of four factors,namely ethanol concentration,solid-liquid ratio,extraction time,and extraction frequency,on the extraction rate of total flavonoids were examined to optimize the best extraction process.The filter paper impregnation method was adopted to assess the antibacterial activity.The outcomes manifested that the linear relationship was favorable when the total flavonoid concentration was within the range of 0.015-0.04 mg/mL(R^(2)=0.9994),the average sample recovery rate was 99.83%(RSD<2%),and both the precision and stability were satisfactory.The optimal extraction process of total flavonoids from Euonymus fortunei was as follows:with 70%ethanol as the extractant,the solid-liquid ratio was 1∶40,and ultrasonic extraction was performed at 60℃twice,90 min each time,with an extraction rate of 17.85%.The antibacterial results indicated that the total flavonoids of Euonymus fortunei exerted a distinct inhibitory effect on Gram-negative bacteria.Within the range of 5-25 mg/mL,the antibacterial effect intensified with the increase in concentration.At the same concentration,the inhibitory effect on different strains was as follows:Escherichia coli>Staphylococcus aureus>Shigella dysenteriae.The optimized extraction process in this study is ecofriendly,convenient,stable and reliable.The extracted total flavonoids have prominent inhibitory effects on Escherichia coli,Staphylococcus aureus and Shigella dysenteriae.
