摘 要:试验旨在探究螺旋藻多糖(PSP)对猪伪狂犬病毒(PRV)感染小鼠脾脏氧化应激的调节作用。将120只BALB/c小鼠随机分5组,分别为高剂量螺旋藻多糖组分1(PSP-1,4 mg)组、中剂量PSP-1(2 mg)组、低剂量PSP-1(1 mg)组、PRV组和空白对照组。高、中、低剂量PSP-1组及PRV组第1天经腹腔注射PRV溶液,空白对照组经腹腔注射灭菌生理盐水。随后1~6 d,高、中、低剂量PSP-1组分别灌胃相应剂量PSP-1,PRV组和空白对照组灌胃灭菌生理盐水,第7天剖杀。采用试剂盒法测定小鼠脾脏丙二醛(MDA)、一氧化氮(NO)含量以及超氧化物歧化酶(SOD)、诱导型一氧化氮合酶(iNOS)酶活性;通过H.E.染色观察小鼠脾脏病理变化;通过免疫组化检测小鼠脾脏中血红素加氧酶-1(HO-1)、iNOS表达。结果显示,与PRV组相比,高剂量PSP-1组小鼠脾脏中MDA、NO含量及iNOS活性极显著降低(P<0.05),SOD活性极显著升高(P<0.05);H.E.染色结果显示,高、中、低剂量PSP-1处理后,小鼠脾脏病理变化有不同程度减轻;免疫组化结果显示,高、中、低剂量PSP-1处理PRV感染小鼠后,脾脏中HO-1的表达有不同程度的升高,iNOS的表达有不同程度的降低。说明PSP-1能够减轻PRV感染小鼠脾脏的损伤,缓解PRV感染小鼠引起的氧化应激。The aim of this experiment was to investigate the regulatory effects of Spirulina platensis polysaccharide(PSP)on oxidative stress in spleen of mice infected with porcine pseudorabies virus(PRV).A total of 120 BALB/c mice were randomly divided into 5 groups,including high-dose PSP-1(4 mg)group,medium-dose PSP-1 group(2 mg)group,low-dose PSP-1 group(1 mg)group,PRV group and blank control group.On the first day,the high,medium,and low-dose PSP-1 groups and the PRV group were intraperitoneally injected with PRV solution,while the blank control group was intraperitoneally injected with sterile physiological saline.Then,from day 1 to 6,the high,medium,and low-dose PSP-1 groups were orally administered with corresponding doses of PSP-1,while the PRV group and blank control group were orally administered with sterile physiological saline.On the 7th day,the mice were sacrificed.The contents of MDA and NO and the enzyme activities of SOD and iNOS in the mouse spleen were deter‐mined by assay kit method.H.E.staining was used to observe the pathological changes of spleen of the animals.The expression of HO-1 and iNOS in the mouse spleen was detected by immunohistochemistry.The findings indicated that the contents of MDA and NO and the activity of iNOS in spleen of mice in high dose PSP-1 group were significantly decreased when compared with PRV group(P<0.05),and the activity of SOD was significantly increased(P<0.05).H.E.staining showed that the pathological changes of spleen were alleviated to different degrees after treatment with different doses of PSP-1.Immunohistochemical results showed that the expression of HO-1 in spleen of PRV-infected mice increased in different doses of PSP-1,while the expression of iNOS decreased in different degrees.The results showed that PSP-1 can alleviate the spleen damage of PRV-infected mice and alleviate oxidative stress caused by PRV-infected mice.
