摘 要:为评价紫苏叶醇提物抗肠炎活性,明确其作为抗生素替代物的应用价值,采用超高效液相色谱-质谱联用(UPLC-MS)法分析紫苏叶醇提物中成分组成,并采用2,4,6-三硝基苯磺酸(TNBS)诱导炎性肠病斑马鱼,考察空白(Control)组、模型(TNBS)组、紫苏叶醇提物(PLAE)组、土霉素+多西环素(OCD)组斑马鱼肠道组织形态、炎性细胞因子[肿瘤坏死因子-α(TNF-α)、白细胞介素6(IL-6)、白细胞介素1β(IL-1β)]的表达情况及免疫指标[免疫球蛋白G(IgG)、免疫球蛋白A(IgA)、免疫球蛋白M(IgM)]水平变化。结果显示:紫苏叶醇提物中共鉴定出阿魏酸、科罗索酸、迷迭香酸、木犀草素、黄芩苷等20种活性成分;紫苏叶醇提物对TNBS诱导的成年斑马鱼肠道组织细胞排列紊乱、肠绒毛脱落具有较好的改善作用,可促进肠道内杯状细胞数量恢复;与TNBS组相比,PLAE组炎性细胞因子TNF-α、IL-6、IL-1β表达量显著降低(P<0.05),免疫指标IgG、IgA、IgM水平显著升高(P<0.05);PLAE组与OCD组间各指标比较均差异不显著(P>0.05)。紫苏叶醇提物中活性成分丰富,具有明显的抗炎、提高免疫力等功效,对斑马鱼炎性肠病的改善具有积极作用,有较好的抗生素替代潜力。To evaluate the anti-enteritis activity of alcohol extract of Perilla frutescens leaves,and to clarify its application value as an antibiotic substitute.Ultra high performance liquid chromatographymass spectrometry(UPLC-MS)was used to analyze the components in the alcohol extract of Perilla leaves,and 2,4,6-trinitrobenzenesulfonic acid(TNBS)was used to induce inflammatory bowel disease in zebrafish.The intestinal tissue morphology,expression of inflammatory cytokines TNF-α,IL-6,IL-1β,and changes in immune indicators IgG,IgA,and IgM levels in the blank group(Control),model group(TNBS),perilla leaf alcohol extract group(PLAE),and oxytetracycline+doxycycline group(OCD)were investigated.The result showed that a total of 20 active ingredients were identified in the alcohol extract of perilla leaves,including ferulic acid,corosolic acid,rosmarinic acid,luteolin,and baicalin;the alcohol extract of perilla leaves can significantly improve the disordered arrangement of intestinal tissue cells and intestinal villus shedding induced by TNBS in adult zebrafish,promote the recovery of goblet cell count in the intestine.Compared with the TNBS group,the expression levels of inflammatory cytokines TNF-α,IL-6,and IL-1βin the PLAE group were significantly reduced(P<0.05),and the levels of immune indicators IgG,IgA,and IgM were significantly increased(P<0.05),there was no statistically significant difference between PLAE group and OCD group(P>0.05).The alcohol extract of perilla leaves is rich in active ingredients and has significant anti-inflammatory and immune enhancing effects.It has a positive effect on the improvement of zebrafish inflammatory bowel disease and has good potential for antibiotic substitution.
