摘  要:目的研究梅州客家人群中载脂蛋白E(ApoE)基因的两个单核苷酸多态性位点(SNP)rs429358和rs7412的分布特征,探讨基因多态性与该人群冠心病易感性的关系。方法选取了梅州地区客家人群86例确诊冠心病患者及同时期185例临床检查无冠心病客观证据的体检人群作为对照组,进行ApoE基因rs429358、rs7412位点的基因型和等位基因检测,分析影响因素在冠心病与非冠心病组间的分布特征、影响关系和对比差异。结果在本次研究人群中,ApoE基因的DNA双链两个SNP位点rs429358和rs7412都存在TT、CC和CT三种类型,rs429358基因型最常见的是TT,rs7412基因型最常见的是CC。在冠心病和非冠心病组中ApoE基因型频率占比最高的都是ε3/ε3,等位基因最常见的都是ε3。等位基因ε2、等位基因ε3在冠心病与非冠心病人群的分布比例不同,存在统计学差异。二元Logistic回归分析显示等位基因ε3为冠心病的保护性因素,回归系数-2.50,比值比为0.08。结论本次研究证明了在梅州地区客家人群中,ApoE基因存在三种等位基因ε2、ε3和ε4,等位基因ε3为冠心病的保护性因素。ObjectiveTo investigates the distribution characteristics of the apolipoprotein E(ApoE)gene′s two single nucleotide polymorphism(SNP)sites,rs429358 and rs7412,within the Hakka population in Meizhou,and to explore the relationship between genetic polymorphisms and susceptibility to coronary artery disease(CAD)within this population.MethodsA total of 86 patients with the diagnosis of CAD from the Hakka population in Meizhou and other 185 check-up individuals without objective evidence of CAD were enrolled.Genetic typing and allele detection of the ApoE gene at rs429358 and rs7412 were conducted.The analysis focused on assessing the distribution characteristics,relationships,and comparative differences of these genetic factors between CAD and non-CAD groups,along with other influencing factors.ResultsAmong the study population,both DNA double-strand SNP sites,rs429358 and rs7412 of the ApoE gene,exhibited three types:TT,CC,and CT.The most prevalent genotype for rs429358 was TT,while CC was the most common for rs7412.The highest frequency of ApoE genotypes in both CAD and non-CAD groups was observed withε3/ε3,and the most common allele wasε3.The distribution proportions ofε2 andε3 alleles differed significantly between CAD and non-CAD groups.Binary logistic regression analysis indicated that theε3 allele serves as a protective factor against CAD,with a regression coefficient of-2.50 and an odds ratio(OR)value of 0.08.ConclusionThis study establishes the existence of three ApoE alleles(ε2,ε3,andε4)within the Hakka population in Meizhou,with theε3 allele identified as a protective factor against CAD.
