摘  要:目的:通过制备大鼠脑缺血再灌注损伤(CIRI)模型探讨针刺对大鼠缺血侧海马组织miR-301a-5p表达的影响。方法:线栓法制备大脑中动脉闭塞/再灌注(MCAO/R)大鼠模型,利用针刺进行干预。采用改良Garcia评分法检测大鼠神经功能,TTC染色法检测脑梗死体积比,RT-qPCR检测缺血侧海马区miR-301a-5p的表达,并利用在线网站数据库TargetScan8.0、mi-RDB及miRWalk预测miR-301a-5p的靶基因,DAVID数据库对靶基因进行基因本体(GO)分析,HE染色观察细胞病理改变。结果:与MCAO/R组相比,MCAO/R+AC干预组大鼠Garcia评分升高(P<0.05),脑梗死体积减小(P<0.01),缺血侧海马区miR-301a-5p表达量显著增加(P<0.01)。预测miR-301a-5p有交集的靶基因101个,GO分析在生物进程(BP)中发现miR-301a-5p靶基因富集于白细胞介素-6(IL-6)的调控及糖皮质激素反应等相关炎症因子,且在MCAO/R组观察到细胞坏死,炎性细胞浸润,MCAO/R+AC组可见散在的炎性细胞浸润。结论:针刺可降低CIRI大鼠神经功能缺损症状,减少脑梗死体积,其机制可能与激活miR-301a-5p的表达抑制细胞炎症有关。Objective:To study the effect of acupuncture on miR-301a-5p expression in the hippocampal tissue of the ischemic side of rats by preparing a rat cerebral ischemia/reperfusion injury( CIRI) rat model.Methods:The middle cerebral artery occlusion/reperfusion( MCAO/R) rat model was established using the thread embolism method and treated with acupuncture as an intervention.The modified Garcia score was used to access neurologic function in rats,and TTC staining was used to measure cerebral infarct volume ratio.RT-q PCR was utilized to detect the expression of miR-301a-5p in the hippocampal region on the ischemic side.The target genes of miR-301a-5p were predicted using online databases such as Target Scan8.0,mi-RDB,and miRWalk.Gene Ontology( GO) analysis of the target genes was conducted via the DAVID database.Additionally,pathological changes in cells were observed using HE staining.Results:Compared with MCAO/R group,Garcia score of rats in MCAO/R + AC group was increased( P < 0.05),and the infarct volume decreased( P < 0.01).The expression of miR-301a-5p in the ischemic hippocampus was significantly increased( P < 0.01).Online database identified 101 intersecting target genes of miR-301a-5p.GO analysis in Biological Processes( BP) revealed that miR-301a-5p target genes were enriched in inflammatory factors,including interleukin-6( IL-6) regulation and glucocorticoid response.In the MCAO/R group,cell necrosis and inflammatory cell infiltration were observed,while scattered inflammatory cell infiltration was noted in the MCAO/R + AC group.Conclusion:Acupuncture alleviates neurological deficit symptoms and decreases cerebral infarction volume in CIRI rats,and the mechanism may be related to the activation of miR-301a-5p expression to suppress cellular inflammation.
