艾燃烧生成物对APP/PS1小鼠认知功能以及海马和内嗅皮层单胺类神经递质的影响

摘  要:目的探究艾燃烧生成物对淀粉样前体蛋白/早老素-1(amyloid precursor protein/presenilin 1,APP/PS1)小鼠海马(hippocampus,HPC)和内嗅皮层(entorhinal cortex,EC)神经元形态和神经递质含量的影响,探讨艾燃烧生成物对阿尔茨海默病的相关作用机制。方法将4月龄雄性APP/PS1小鼠随机分为模型组、艾燃烧生成物组、精油组,设同月龄雄性C57BL/6小鼠作为空白组,每组9只。艾燃烧生成物组与精油组分别进行10~15 mg/m 3的艾燃烧生成物和1%迷迭香精油吸嗅干预,空白组和模型组仅抓取固定。所有干预每日1次,每次20分钟,每周6次,共计12周。实验结束后,采用新物体识别实验检测各组小鼠认知功能,通过HE染色观察各组小鼠HPC和EC的神经元形态,采用ELISA法检测HPC和EC多巴胺(dopamine,DA)、5-羟色胺(5-hydroxytryptamine,5-HT)、去甲肾上腺素(norepinephrine,NE)含量。结果在新物体识别实验中,与模型组比,艾燃烧生成物组和精油组的新物体认知指数和区分指数均提高(P<0.01);HE染色法:与模型组比,艾燃烧生成物组和精油组小鼠HPC和EC的神经元形态得到改善;ELISA法:与模型组相比,精油组小鼠HPC和EC两区域DA、5-HT及NE含量均升高(P<0.05),艾燃烧生成物组小鼠HPC和EC两区域5-HT、NE和DA含量均升高(P<0.05)。结论艾燃烧生成物干预可以缓解APP/PS1小鼠HPC和EC两区域神经元形态损伤,提高两区域5-HT、NE和DA含量,从而改善其认知功能。Objective To investigate the effects of moxa combustion products intervention on the neuronal morphology and neurotransmitter content in the hippocampus(HPC)and entorhinal cortex(EC)of amyloid precursor protein/presenilin 1(APP/PS1)mice,and to explore the mechanisms of moxa combustion products on Alzheimer’s disease.Methods 4-month-old male APP/PS1 mice were randomly divided into the model group,the moxa combustion products group,and the essential oil group,and male C57BL/6 mice of the same age were set up as the control group,with 9 mice in each group.Mice in the essential oil group was given 1%rosemary essential oil for sniffing.Mice in the moxa combustion products group received an exposure of 10~15 mg/m 3 of moxa combustion products.Mice in control and the model groups were captured and fixed daily without any intervention.All interventions were conducted once a day for 20 minutes,6 days a week for 12 weeks.At the end of the experiment,the learning memory level of mice in each group was detected using the novel object recognition test,the morphology of neurons in the HPC and EC regions of mice in each group was observed using HE staining,and dopamine(DA),5-hydroxytryptamine(5-HT),and norepinephrine(NE)levels in the two regions were measured using ELISA.Results In the novel object recognition experiment,mice in the moxa combustion products group and the essential oil group had increased indices of new object recognition and discrimination compared with the model moxa combustion products group(P<0.01);the result of HE staining showed that,there was an improvement in neuronal form of HPC and EC in mice in the moxa combustion products group and essential oil group compared to the model group;the result of ELISA showed that,both the DA,5-HT and NE levels in the HPC and EC regions of mice in the essential oil group were elevated compared with the model group(P<0.05),and the 5-HT,NE and DA levels in both the HPC and EC regions of mice in the moxa combustion products group were elevated(P<0.05).Conclusion Moxa comb
