槐耳清膏对膀胱癌细胞增殖和迁移的影响

摘　　要：目的 研究槐耳清膏对膀胱癌细胞增殖和迁移的影响。方法 使用CCK-8法、平板克隆及Transwell实验检测膀胱癌细胞的增殖、侵袭与迁移能力。通过Western blot法检测膀胱癌细胞基质金属蛋白酶-2(MMP-2)、基质金属蛋白酶-9(MMP-9)、波形蛋白(Vimentin)、N-钙黏蛋白(N-cadherin)、E-钙黏蛋白(E-cadherin)的表达水平。结果 CCK-8实验结果表明,采用低浓度组(3 mg/ml)、中浓度组(6 mg/ml)、高浓度组(12 mg/ml)的槐耳清膏分别处理人膀胱癌细胞系RT4和鼠膀胱癌细胞系MB49,24、48、72 h后细胞活力被显著抑制(P<0.05),且呈浓度和时间依赖性。增殖侵袭实验结果表明,与对照组相比,低、中、高浓度的槐耳清膏显著抑制膀胱癌细胞增殖、侵袭迁移能力(P<0.05)。Western blot结果表明,与对照组相比,低、中、高浓度组细胞中的MMP-2、MMP-9、Vimentin、N-cadherin表达水平降低,E-cadherin蛋白表达水平升高,差异具有统计学意义(P<0.05)。结论 槐耳清膏通过改变膀胱癌细胞上皮-间质转化相关蛋白水平的表达,抑制膀胱癌细胞的增殖、迁移与侵袭。Objective To investigate the effect of Huaier aqueous extract on the proliferation and migration of bladder cancer cells.Methods CCK-8 method and plate cloning were applied to detect the proliferation ability of bladder cancer cells,and Transwell assay was used to detect the invasive and migratory ability of bladder cancer cells.Western blot was used to detect the expression levels of matrix metalloproteinase-2(MMP-2),matrix metalloproteinase-9(MMP-9),Vimentin,Ncadherin and E-cadherin in bladder cancer cells.Results The results of CCK-8 showed that the cell viability of RT4 and MB49 cells was significantly inhibited in a concentration and time dependent manner after treating with low concentration(3 mg/ml),the middle concentration(6 mg/ml),and high concentration(12 mg/ml),of Huaier aqueous extract,respectively,for 24、48、72 h( P<0.05).The results of proliferation-invasion assay showed that compared with the control group,the proliferation,invasion and migration ability of bladder cancer cells were reduced by the low,middle and high concentrations of Huaier aqueous extract(P<0.05).The results of Western blot showed that compared with the control group,the expression levels of MMP-2,MMP-9,Vimentin,and Ncadherin reduced,while the expression level of E-cadherin protein increased in the cells of the low,middle and high concentration groups.There was a significant difference between the groups(P <0.05).Conclusions Huaier aqueous extract can inhibit bladder cancer cells proliferation,migration,and invasion by altering the expression of epithelial mesenchymal transition-related protein levels in bladder cancer cells.
