摘 要:目的:通过建立酒精联合高脂饮食诱导大鼠酒精性脂肪肝动物模型,探讨黄甲软肝颗粒对大鼠血清脂质和肝功能指标的影响。方法:SD大鼠60只,除10只正常组以正常饲料喂养,其余大鼠喂养高脂饲料并灌胃30%的酒精8 mL/kg造模。酒精联合高脂饮食喂养两周后通过眼眶取血检测血脂指标,将造模成功大鼠按总胆固醇(CHO)含量随机分成6组。分组后,正常组继续用正常饲料喂养,饮用蒸馏水,给药时灌胃给予蒸馏水;除正常组外其余各组用高脂饲料喂养,上午统一灌胃给予30%酒精8mL/kg,下午模型组灌胃给予蒸馏水10mL/kg,其余组分别灌胃给予相应药物,每天给药1次,连续给药8周,末次给药后禁食进行各指标检测。结果:①黄甲软肝中高剂量组CHO和LDL-C含量降低(P>0.05),TG含量显著降低;②黄甲软肝颗粒中剂量组大鼠血清ALT、AST、ALP活性显著降低(P<0.05或P<0.01);③黄甲软肝颗粒中、高剂量组和阳性对照药组大鼠血清MDA含量显著降低(P<0.05),SOD活性显著增加(P<0.05);④黄甲软肝颗粒中、高剂量组大鼠肝细胞结构有明显改善,体积有较明显减小,脂滴数量明显减少。结论:黄甲软肝颗粒能显著降低血清中CHO、TG和LDL-C的水平,降低肝功能指标ALT、AST和ALP的活性,改善肝脏的组织病理学状态,对酒精联合高脂饮食诱导酒精性脂肪肝有较好的作用。Objective To investigate the effects of Huangjia Ruangan Granules on serum lipids and liver function indexes in rats by establishing an animal model of alcoholic fatty liver induced by alcohol+high-fat diet.Methods Seventy SD rats were fed a high-fat diet and gavaged with 30%alcohol at 8 mL/kg,except for 10 normal rats in the control group,which were fed a normal diet.After two weeks of alcohol+high-fat diet,the rats were randomly divided into 6 groups according to the total cholesterol(CHO)content by orbital blood sampling for the detection of lipid indexes.After grouping,the normal control group continued to be fed with normal feed,drinking distilled water,and giving distilled water by gavage during drug administration;the remaining groups except the normal control group were fed with high-fat diet,and 30%alcohol was given by gavage for 8 mL/kg in the morning,distilled water was given by gavage for 10 mL/kg in the afternoon in the model control group,and the remaining groups were respectively given the corresponding drugs by gavage,and the drugs were given once a day for 8 w,and then fasted for each index test after the last administration.After the last administration of the drug,fasting was carried out for the detection of various indexes.Results①CHO and LDL-C content decreased in the middle and high dose groups of Huangjia Ruangan Granules(P>0.05),and TG content decreased significantly;②serum ALT,AST and ALP activities decreased significantly in the middle dose group of Huangjia Ruangan Granules(P<0.05 or P<0.01);③serum MDA content decreased significantly in the middle and high dose groups of Huangjia Ruangan Granules and in the positive control group(P<0.05),and SDA content decreased significantly in the middle and high dose groups of Huangjia Ruangan Granules and the positive control group(P<0.05),SOD activity increased significantly(P<0.05);④Huangjia Ruangan Granules medium and high dose group of rats hepatocyte structure has been significantly improved,the volume has been reduced significantly,th
