柴胡疏肝汤对癫痫大鼠IL-1β、IL-18表达的影响

摘  要:目的:探讨柴胡疏肝汤对癫痫大鼠脑组织白细胞介素-1β(IL-1β),白细胞介素-18(IL-18)表达的影响。方法:将SD大鼠分为空白组、模型组、柴胡疏肝汤低剂量治疗组、柴胡疏肝汤中剂量治疗组和柴胡疏肝汤高剂量治疗组,采用腹腔注射氯化锂溶液联合盐酸匹罗卡品诱发癫痫大鼠模型,成功建立癫痫大鼠模型48只后,将其随机分为4组,每组各12只;其中柴胡疏肝汤低剂量治疗组、柴胡疏肝汤中剂量治疗组、柴胡疏肝汤高剂量治疗组采用相应剂量的柴胡疏肝汤干预,空白组、模型组采用等量的生理盐水干预,每天1次,连续4周。末次给药后,观察各组大鼠癫痫发作情况,记录癫痫发作频率、持续时间及发作等级,然后每组大鼠断头取脑,分离海马组织,通过反转录-聚合酶链反应(RT-PCR)及蛋白免疫印迹(Western Blot)检测大鼠海马组织中IL-1β和IL-18 mRNA及蛋白的表达。结果:柴胡疏肝汤治疗组癫痫大鼠经治疗后,癫痫发作时间和发作级别均明显下降,而模型组癫痫大鼠治疗前后无明显变化;与模型组相比,柴胡疏肝汤治疗组大鼠海马组织中IL-1β和IL-18 mRNA及蛋白的表达均降低(P<0.05),且高剂量的柴胡疏肝汤对癫痫大鼠的IL-1β和IL-18 mRNA及蛋白表达下降最明显(P<0.05)。结论:柴胡疏肝汤可下调癫痫大鼠海马组织中IL-1β和IL-18的表达,减少癫痫大鼠的痫性发作。Objective To investigate the effect of Chaihu Shugan Decoction on the expression of interleukin-1β(IL-1β)and interleukin-18(IL-18)in brain tissue of epileptic rats.Methods SD rats were divided into blank group,model group,Chaihu Shugan Decoction low-dose treatment group,Chaihu Shugan decoction medium-dose treatment group,Chaihu Shugan decoction high-dose treatment group,The epileptic model was induced by intraperitoneal injection of lithium chloride solution combined with pilocarpine hydrochloride.After the epilepsy model was established,the successful rats were selected and grouped.Buplehu Shugan Decoction low dose treatment group,Buplehu Shugan decoction medium dose treatment group,Buplehu Shugan Decoction high dose treatment group were treated with corresponding dose of buplehu Shugan decoction intervention,blank group and model group were treated with equal amount of normal saline intervention.The mRNA and protein expression of IL-1βand IL-18 in rat hippocampus were detected by reverse transcription-polymerase chain reaction(RT-PCR)and Western Blot.Results 1.The duration and grade of seizures of epileptic rats in the treatment group of Chaihu Shugan Decoction decreased significantly after treatment,while there was no significant change before and after treatment of epileptic rats in the model group.2.Compared with the model group,the expressions of IL-1βand IL-18 mRNA and protein in hippocampal tissue of rats in the treatment group were decreased(P<0.05),and the expression of IL-1βand IL-18 mRNA and protein in epileptic rats was most significantly decreased by high dose of Chaihu Shugan Decoction(P<0.05).Conclusion Chaihu Shugan Decoction can down-regulate the expression of IL-1βand IL-18 in hippocampal tissue of epileptic rats and reduce epileptic seizures in epileptic rats.
