摘  要:灰盖粉褶鹅膏Amanita griseorosea是鹅膏属檐托鹅膏组的一种剧毒鹅膏,主要分布于中国华南地区,其子实体内所含的毒素种类、含量,及编码基因尚待深入研究。通过甲醇提取、液质联用LC-MS检测方法对其毒素种类和含量进行检测,结果显示在该鹅膏物种中仅检测到了α-amanitin,其平均含量为(2 815.70±835.93)μg/g。通过MSDIN家族基因简并引物对灰盖粉褶鹅膏环肽编码基因进行PCR扩增及克隆测序,获得了7条环肽编码基因序列。序列比对及聚类分析结果显示,其中1条编码α-amanitin,1条编码已报道的未知环肽,其余5条均编码未报道的新环肽。研究结果为后期深入了解灰盖粉褶鹅膏毒性、环肽合成能力及挖掘更多新的环肽奠定了重要基础。Amanita griseorosea is a lethal Amanita species within the sect.Phalloideae,mainly distributing in South China.In this study,toxins were detected through the methanol extraction and LC-MS analysis.Results showed that onlyα-amanitin was measured from this species,revealing an average concentration of(2815.70±835.93)μg/g.In addition,genomic fragments from the MSDIN gene family were amplified by PCR for cloning and sequencing using degenerate primers.Seven MSDIN genes encoding different cyclic peptides were obtained.Sequence comparison and phylogenetic analysis indicated that one encodes theα-amanitin and another one encodes a cyclic peptide as previous reported.The remaining five all encode novel cyclic peptides.The above results thereafter provide an important foundation for further deep understanding of the toxicity and the capacity of cyclic peptide biosynthesis as well as the exploration of new cyclic peptides in A.griseorosea.
