摘 要:目的构建日光性皮炎小鼠瘙痒模型,检测小鼠瘙痒及焦虑、抑郁样行为,并观察内侧前额叶皮层(mPFC)和前扣带回皮层(ACC)内神经元的激活情况。方法通过宽幅紫外线照射构建日光性皮炎小鼠模型。利用HE染色技术观察小鼠颈背部皮肤的病理变化。通过搔抓实验、高架十字迷宫、旷场和悬尾实验进行痒行为、焦虑和抑郁样行为检测。采用免疫荧光染色技术来观察mPFC和ACC内神经元的活性变化。结果HE染色结果表明,对照组小鼠皮肤组织呈现出正常的形态结构,模型组小鼠背部皮肤可见明显红斑,棘细胞层出现特征性的“晒斑细胞”。与对照组相比,模型组小鼠搔抓次数显著增多(P<0.01);在旷场实验中的中央区域探索时间减少(P<0.05);在高架十字迷宫实验中的开臂探索时间缩短(P<0.01);在悬尾实验中的不动时间增加(P<0.05)。免疫荧光染色显示,模型组小鼠mPFC和ACC脑区FOS阳性细胞数量显著增加(P<0.01)。结论成功建立了日光性皮炎瘙痒模型,并观察到模型小鼠伴发焦虑和抑郁样行为。模型组小鼠mPFC和ACC脑区内神经元激活,上述结果为mPFC和ACC参与日光性皮炎瘙痒及其伴发的负性情绪提供了形态学证据。Objective To construct a mouse model of solar dermatitis,detect itch and anxiety and depression-like behaviors in mice,and observe the activation of neurons in the medial prefrontal cortex(mPFC)and anterior cingulate cortex(ACC).Methods A mouse model of solar dermatitis was constructed by broad-band ultraviolet B irradiation.The pathological changes of the skin of the neck and back of mice were observed by HE staining.Itch behavior and anxiety and depression-like behavior were detected by scratch test,elevated plus maze test,open field test and tail suspension test.The activation of neurons in mPFC and ACC was observed using immunofluorescence staining.Results The results of HE staining showed that the skin tissue of mice in the control group showed normal morphological structure,while the back skin of mice in the model group showed obvious erythema and characteristic“sunburned cells”in the acanthocyte layer.Compared with the control group,the model group showed a significant increase in the scratching bouts(P<0.01),a reduction in the exploration time in the central area in the open field test(P<0.05)and in the exploration time in the open arm in the elevated plus maze test(P<0.01),and an increase in the immobilization time in the tail suspension test(P<0.05).Immunofluorescence staining revealed a significant increase in the number of FOS positive cells in mPFC and ACC in the model group(P<0.01).Conclusion In this study,we successfully established itch model of solar dermatitis and observed anxiety and depression-like behaviors in the model mice.Some neurons in mPFC and ACC were activated in the model group.The study provides morphological evidence for the involvement of mPFC and ACC in solar dermatitis itch and its accompanying negative emotions.
