摘  要:目的:建立超高效液相色谱串联质谱法(UPLC-MS/MS)同时测定沙苑子中11种黄酮类成分(杨梅素-3-O-β-D-葡萄糖苷、沙苑子苷A、沙苑子苷B、紫云英苷、毛蕊异黄酮葡萄糖苷、鼠李柠檬素-3-O-β-D-葡萄糖苷、毛蕊异黄酮、异槲皮苷、番石榴苷、芒柄花素、鼠李柠檬素)的含量。方法:采用Waters ACQUITY UPLC BEH C_(18)柱(100 mm×2.1 mm,1.7μm);以甲醇和0.1%甲酸水溶液为流动相,梯度洗脱,流速0.2 mL/min;柱温40℃;进样量为1μL;电喷雾离子化正、负离子模式,多反应监测模式。结果:在考察的浓度范围内,11种黄酮类成分线性关系良好(r≥0.99955);平均加样回收率和RSD分别为97.63%~102.87%和0.96%~2.73%。结论:该方法准确度、精密度、稳定性、线性关系良好,可用于沙苑子中11种黄酮类成分的含量测定。Objective:To establish a method for simultaneous determination of 11 flavonoid components(myricetin-3-O-β-Dglucopyranoside,complanatoside A,complanatoside B,astragalin,calycosin-7-O-β-D-glucoside,rhamnocitrin 3-glucoside,calycosin,isoquercitrin,guaijaverin,formononetin,rhamnocitrin)in Astragali Complanati Semen by ultra high performance liquid chromatography tandem mass spectrometry(UPLC-MS/MS).Methods:Using Waters ACQUITY UPLC BEH C_(18) column(100 mm×2.1 mm,1.7μm),methanol and 0.1%formic acid aqueous solution as mobile phases,gradient elution was performed at a flow rate of 0.2 mL/min.Column temperature was 40℃and injection volume was 1μL.The positive and negative ion modes and the multi reaction monitoring mode were adopted for the electro spray ionization.Results:Within the concentration range under investigation,there was a good linear relationship(r≥0.99955)among the 11 flavonoid components;The average sample recovery rate and RSD were ranged from 97.63%-102.87%and 0.96%-2.73%.Conclusion:This method has good accuracy,precision,stability,and linear relationship,and can be used for the determination of eleven flavonoid components in Astragali Complanati Semen.
