摘  要:【目的】2023年11月在湖南省宜章县金刚藤上发现一种锈病发生严重,为探究金刚藤上锈病的病原,开展金刚藤锈病病原鉴定相关研究。【方法】光学显微镜下观察患病叶片上病原菌的形态,然后取金刚藤叶片发病部位提取总DNA,用通用引物扩增病原菌ITS和LSU序列,并切胶回收扩增片段,连接T载体测序,测序结果NCBI数据库比对,并构建系统发育树。【结果】湖南省宜章县金刚藤上发现的病害是由柄锈菌属(Puccinia sp.)真菌引起。【意义】明确了湖南省宜章县金刚藤病害的病原菌,为今后防控工作的开展提供了参考。[Objective]In November 2023,a leaf rust disease was discovered on Smilax china in Yizhang County,Hunan Province.To investigate the cause of the disease,relevant researches were conducted.[Method]The morphology of the diseased part of the leaves was observed under the optical microscope,and then the total DNA of the diseased part of the leaves was extracted,and the ITS and LSU sequences of pathogen were amplified with universal primers.The amplified fragments were recovered by cutting the gel,and sequenced.The sequencing results were compared with the NCBI database and phylogenetic tree was conducted.[Result]The pathogen of rust fungus on Smilax china was identified as Puccinia sp..[Significance]The study provided references for the prevention and control of the disease in the future.
