摘　　要：建立了超高效液相色谱-串联质谱法同时测定干血斑中维生素A和维生素E的含量。样品经纯化水超声提取,甲醇沉淀蛋白,乙酸乙酯萃取,上层萃取液氮气吹干后复溶。采用Accucore C_(18)色谱柱(50 mm×2.1 mm,2.6μm)分离,在正离子多反应监测模式下进行检测。维生素A、维生素E的质量浓度分别在0.1~3、0.5~30 mg/L范围内和相应色谱峰面积与同位素内标色谱峰面积之比的线性关系良好,相关系数均大于0.995。维生素A、维生素E的样品加标回收率分别为87.2%~92.8%、98.6%~108.5%,测定结果的相对标准偏差均小于8%(n=6)。该方法适用于干血斑中维生素A和维生素E的同时测定。A method for simultaneous determination of vitamin A and vitamin E in dried blood spots(DBS)by ultrahigh performance liquid chromatography tandem mass spectrometry was established.The sample was purified by ultrasonic extraction with water,protein was precipitated with methanol,extracted with ethyl acetate,and the upper extract was dried with nitrogen and then dissolved again.Separation was performed using an Accucore C_(18) column(50 mm×2.1 mm,2.6μm)and detection was carried out in positive ion multi reaction monitoring mode.Vitamin A and vitamin E had good linear relationships with the ratio of the peak areas of the corresponding chromatographic peak to the peak areas of the isotopic internal standard in the mass concentration range of 0.1-3 mg/L and 0.5-30 mg/L,respectively.The correlation coefficients were all more than 0.995.The recoveries were 87.2%-92.8%and 98.6%-108.5%for vitamin A and vitamin E.The relative standard deviations of repeatable tests were less than 8%(n=6).The method was suitable for the simultaneous determination of vitamin A and vitamin E in DBS.
