摘 要:利用植物DORN1突变体(dorn1-3,其中T-DNA破坏了植物细胞外ATP受体DORN1编码基因),研究在盐胁迫下DORN1突变对拟南芥种子萌发及H_(2)O_(2)代谢的影响.结果表明,在盐胁迫下DORN1突变能够显著降低种子的萌发.盐胁迫显著提高了植物种子萌发中的H_(2)O_(2)含量和H_(2)O_(2)清除酶的活性;且H_(2)O_(2)的淬灭剂能够缓解盐胁迫对植物种子萌发的抑制,表明盐胁迫下种子萌发的下降和H_(2)O_(2)代谢密切相关.DORN1突变能够导致种子萌发中H_(2)O_(2)含量和H_(2)O_(2)清除酶活性的显著提高提示DORN1突变可能通过改变H_(2)O_(2)代谢影响盐胁迫下的种子萌发,表明在盐压力下细胞外ATP受体DORN1对拟南芥种子萌发具有重要的调节作用,且该作用与H_(2)O_(2)代谢有关.The effects of DORN1 mutation on the germination and H_(2)O_(2) metabolism of Arabidopsis thaliana seeds under salt stress were studied using DORN1 mutant(dorn1-3,in which T-DNA destroys the gene encoding exATP receptor DORN1).The results showed that DORN1 mutation could significantly reduce seed germination under salt stress,for the latter significantly increased the content of H_(2)O_(2) and the activities of H_(2)O_(2)-scavenging enzymes in seed germination.The quenching agent of H_(2)O_(2) could alleviate the inhibition of salt stress on the seed germination,indicating that a decrease of seed germination under salt stress could be closely related to H_(2)O_(2) metabolism.Further observation showed that under salt stress DORN1 mutation could significantly increase the content of H_(2)O_(2) and the activities of H_(2)O_(2)-scavenging enzymes during seed germination,suggesting that DORN1 mutation might affect seed germination under salt stress by altering the metabolism of H_(2)O_(2).These observations suggest that the exATP receptor DORN1 plays an important regulatory role in Arabidopsis seed germination under salt stress,and this role is related to H_(2)O_(2) metabolism.
