摘  要:目的 研究芦荟大黄素(AE)在体外对Cu^(2+)存在下Aβ_(42)聚集及细胞毒性的抑制作用。方法 实验设置Aβ_(42)组、(Aβ_(42)+Cu^(2+))组、(Aβ_(42)+AE)组、(Aβ_(42)+Cu^(2+)+AE)组,应用硫黄素T荧光实验、浊度法、透射电镜及CCK8细胞毒性实验分别探究各组Aβ_(42)的纤维化聚集过程、总聚集程度、聚集体形貌及细胞毒性。结果 与空白组相比,Cu^(2+)可增强Aβ_(42)的总聚集程度,诱导Aβ_(42)聚集形成低β-折叠结构的无规则聚集体,显著降低人神经母细胞瘤细胞SH-SY5Y的活力。AE不仅能抑制Aβ_(42)自发聚集,而且能抑制Cu^(2+)诱导的Aβ_(42)聚集。与(Aβ_(42)+Cu^(2+))组相比,AE能降低Aβ_(42)的总聚集程度,诱导Aβ_(42)形成低β-折叠结构的近圆形或短粗棒状聚集体,并提高细胞活力。结论 AE可抑制Cu^(2+)存在下Aβ_(42)聚集及细胞毒性。Objective To determine the inhibitory effect of aloe emodin (AE) on the aggregation and cytotoxicity of Aβ_(42) in the presence of Cu^(2+)in vitro.Methods An Aβ_(42) group,a (Aβ_(42)+Cu^(2+)) group,a (Aβ_(42)+AE) group,and a (Aβ_(42)+Cu^(2+) +AE) group were set up in the experiment.The fibrotic aggregation process,total aggregation degree,aggregates morphology and cytotoxicity of Aβ_(42) in each group were studied by thioflavin T fluorescence test,turbidity test,transmission electron microscopy test,and CCK8 cytotoxicity test.Results Compared with the blank group,Cu^(2+)enhanced the total degree of Aβ_(42) aggregation,induced the Aβ_(42) aggregation to form irregular aggregates with low β-sheet structure,and greatly reduced the viability of human neuroblastoma cells SH-SY5Y.AE inhibited not only auto aggregation of Aβ_(42),but also Cu^(2+)-induced aggregation of Aβ_(42).Compared with the (Aβ_(42)+Cu^(2+)) group,AE reduced the total aggregation degree of Aβ_(42),induced Aβ_(42) to form near-round or short rod-like aggregates with low β-sheet structure,and increased the cell viability.Conclusion AE can inhibit Aβ_(42) aggregation and cytotoxicity in the presence of Cu^(2+).
