摘 要:目的 建立柴胡牡蛎颗粒中6种成分的一测多评(QAMS)定量分析方法。方法 采用Waters C18反相色谱柱;流动相为乙腈-0.5%磷酸溶液,梯度洗脱;流速为0.8 mL·min^(-1);柱温为25℃,检测波长为274 nm。以黄芩苷为内参物,建立HPLC一测多评法测定黄芩苷、野黄芩苷、千层纸素A-7-O-β-D-葡萄糖醛酸苷、汉黄芩苷、黄芩素、汉黄芩素的含量。结果 6种成分在各自的浓度范围内线性关系良好(r≥0.999)。以黄芩苷为内参物,野黄芩苷、千层纸素A-7-O-β-D-葡萄糖醛酸苷、汉黄芩苷、黄芩素和汉黄芩素的相对校正因子分别为1.80、1.14、0.91、0.69、0.62。7批柴胡牡蛎颗粒中以上6种成分的含量分别为7.80~8.61、0.32~0.36、0.94~1.09、1.77~1.83、0.17~0.18、0.12~0.13 mg·g^(-1)。结论 建立的QAMS法简便、经济、适用性好,可为柴胡牡蛎颗粒的质量控制研究提供基础。Objective To establish a quantitative analysis of multi-components by single marker (QAMS) to determine 6 components in Chaihu Muli granules.Methods Waters C18 reverse chromatographic column was used.The mobile phase was acetonitrile-0.5% phosphoric acid solution with gradient elution;the flow rate was 0.8 mL·min^(-1);the column temperature was 25℃;and the detection wavelength was 274 nm.Baicalin was used as the internal standard,a QAMS method was established to simultaneously determine the content of baicalin,scutellarin,oroxylin A-7-O-β-D-glucuronide,wogonoside,baicalein and wogonin.Results The 6 components showed good linearity in their respective concentration ranges (r≥0.999).With baicalin as the internal standard,the relative correction factors of scutellarin,oroxylin A-7-O-β-D-glucuronide,wogonoside,baicalein and wogonin were 1.80,1.14,0.91,0.69 and 0.62,respectively.The 6 components in 7 batches of Chaihu Muli granules were 7.80~8.61,0.32~0.36,0.94~1.09,1.77~1.83,0.17~0.18,and 0.12~0.13 mg·g^(-1).Conclusion The QAMS method is simple,economical and feasible,which provides a basis for the quality control of Chaihu Muli granules.
