摘　　要：目的建立无糖型胸痛颗粒含量测定方法。方法采用高效液相色谱法测定无糖型胸痛颗粒中丹酚酸B的含量,样品用80%甲醇超声提取30min。采用Agilent1260色谱仪,KromasilC18色谱柱(4.6mm×250mm,5μm);流动相:甲醇-乙腈-甲酸-水(30∶10∶1∶59);检测波长:286nm;柱温:25℃;流速:1.0ml·min^(-1)。结果丹酚酸B进样量在0.40~1.40μg范围内与其峰面积有良好的线性关系,相关系数r=0.9998,该法平均回收率为102.30%,RSD为1.98%(n=6),三批无糖型胸痛颗粒中丹酚酸B平均含量为3.27mg·g^(-1)。结论本法稳定可靠,重现性好,可为无糖型胸痛颗粒质量标准的建立提供参考。Objective To establish a method for quantitative determination of Xiongtong granules(sugar free).Methods High performance liquid chromatography was used to determine the content of salvianolic acid B in Xiongtong granules(sugar free).The sample was extracted with 80%methanol under ultrasonic management for 30 mins.Chromatographic conditions involved Agilent 1260,Kromasil C18(4.6 mm×250 mm,5μm);a mobile phase composed of methanol-acetonitrile-formic acid-water(30∶10∶1∶59),a velocity of 1.0 ml·min^(-1),a detecting wavelength of 286 nm,and a column temperature of 25℃.Results Within the range of 0.40 to 1.40μg,the calibration cure was linear(r=0.9998).The mean recovery rate was 102.30%,RSD 1.98%.The mean content of salvianolic acid B in the three batches of medicine was 3.27 mg·g^(-1).Conclusion This determination method is highly stable,accurate,reproducible,and can be used as a quality standard for Xiongtong granules(sugar free).
