摘 要:目的:对在万古霉素进行血药浓度检测中应用高效液相色谱仪优化进行研究。方法:将含分析有万古霉素的血浆样本放置在剂量为100μL、浓度为10%的高氯酸-甲醇-乙腈混合溶液进行沉淀,比例为2:1:1;而后对其进行离心处理,并选取剂量为150μL的上清液,而后需要将上述液体放置在内衬管内,并且需要将流动相以乙腈一水进行干预,同时将流速保持在0.8mL/min,色谱柱为AgilentZORBAXSB-C18,将柱温保持在40˚C,检测波长为220nm,对其进行分析。对该方法的各项情况进行分析。结果:当万古霉素血药浓度保持在如下范围内的线性关系良好,即:1.18~75.80μg/mL,R2=0.9994;日内、日间精密度值均在15%以下,万古霉素在低、中、高三个浓度下在室温、4˚C、-20˚C环境下的稳定性研究表明,其相对标准偏差(RSD)均在15%以下,显示其在不同温度条件下具有较好的稳定性。结论:优化高效液相色谱法可以有效测定万古霉素的血药浓度,值得临床应用。Objective:To optimize the application of high-performance liquid chromatography in the detection of vancomycin blood drug concentration.Methods:Plasma samples containing vancomycin were placed in a 10%perchloric acid methanol acetonitrile mixed solution at a dose of 100μL for precipitation,with a ratio of 2:1:1;Then centrifuge it and select a supernatant with a dose of 150μL.The above liquid needs to be placed in an inner liner,and the mobile phase needs to be intervened with acetonitrile water,while maintaining a flow rate of 0.8mL/min.The chromatographic column was Agilent ZORBAX SB-C18,and the column temperature is maintained at 40˚C.The detection wavelength was 220nm,and it needs to be analyzed.Analyze the various aspects of this method.Results:The linear relationship was good when the blood concentration of vancomycin was maintained within the range of 1.18-75.80μg/mL,R2=0.9994;The intra-and inter day precision values were all below 15%.Stability studies of vancomycin at low,medium,and high concentrations at room temperature,4˚C,and-20˚C showed that its relative standard deviation(RSD)was below 15%,indicating good stability under different temperature conditions.Conclusion:Optimizing high-performance liquid chromatography can effectively determine the blood concentration of vancomycin and is worthy of clinical application.
