麻杏石甘汤干预流感病毒肺炎小鼠前后肺部菌群变化的研究

摘  要:目的探究小鼠感染流感病毒肺炎前后肺部菌群的变化及经麻杏石甘汤干预后对小鼠肺部菌群的影响。方法采取滴鼻造模的方法建立流感病毒肺炎小鼠模型,经麻杏石甘汤或生理盐水干预不同时间后,提取小鼠肺组织进行16S rDNA测序数据处理及个性分析,探究麻杏石甘汤对流感病毒肺炎小鼠肺部菌群的影响。结果16S rDNA测序结果显示,模型组厚壁菌门、拟杆菌门相对丰度降低,变形菌门、泛菌属、志贺菌属相对丰度增加。麻杏石甘汤组和奥司他韦组能降低变形菌门、志贺菌属相对丰度,增加厚壁菌门相对丰度。Alpha多样性分析显示给药7 d后麻杏石甘汤组、奥司他韦组小鼠肺部菌群丰富度及多样性均较模型组增加(P<0.05);Beta多样性分析结果显示,给药7 d后,除模型组外各组样品点之间的距离小于给药3 d后,组间肺菌群构成有差异。结论麻杏石甘汤干预流感病毒肺炎小鼠后,抑制小鼠肺部菌群中的有害菌增长,减少肺组织损伤程度。Objective To investigate the changes in lung microbiota in mice before and after infection with influenza virus pneumonia,and the effect of intervention with Maxing Shigan Decoction(麻杏石甘汤)on lung microbiota in mice.Methods A mouse model of influenza virus pneumonia was established by nasal drip modeling.After intervention with Maxing Shigan Decoction or physiological saline for different times,mouse lung tissue was extracted for 16S rDNA sequencing data processing and personality analysis,to explore the effect of Maxing Shigan Decoction on the lung microbiota of influenza virus pneumonia mice.Results The 16S rDNA sequencing results showed that the relative abundance of Firmicutes and Bacteroidetes decreased in the model group,while the relative abundance of Proteobacteria,Pantobacteria,and Shigella increased.The Maxing Shigan Decoction group and the oseltamivir group can reduce the relative abundance of Proteobacteria and Shigella,and increase the relative abundance of Firmicutes.Alpha diversity analysis showed that after 7 days of administration,the richness and diversity of lung microbiota in the Maxing Shigan Decoction group and the oseltamivir group of mice increased compared to the model group(P<0.05);The Beta diversity analysis results showed that after 7 days of administration,the distance between sample points in each group except for the model group was less than 3 days after administration,and there were differences in the composition of lung microbiota between the groups.Conclusion After intervening with influenza virus pneumonia in mice,Maxing Shigan Decoction promotes the growth of beneficial bacteria in the lung microbiota of mice and reduces the degree of lung tissue damage.
