摘 要:目的:优化黑根霉胞外多糖(EPS)发酵条件,以获得黑根霉EPS发酵最优工艺,为后续药理学研究和药物开发提供材料保障。方法:响应面法(RSM)考察前段发酵时间(变温时间)、后段发酵温度和发酵时间3个因素对EPS产量的影响,预测EPS分批发酵最优工艺。结果:RSM结果显示,各因素按照对响应值的影响顺序为:发酵时间>后段发酵温度>前段发酵时间。发酵时间(P<0.001)、后段发酵温度(P=0.015)对EPS产量的影响显著。预测EPS发酵最优工艺为:前段发酵时间58.018 h,后段发酵温度26.984℃,发酵时间143.193 h,EPS产量为0.891 g/L。为验证工艺可靠性,结合设备条件和控制能力,将验证工艺条件优化设置为:前段发酵时间58 h,后段发酵温度27℃,发酵时间143 h。经验证,EPS产量为0.889 g/L。结论:通过RSM优化工艺,可以获得黑根霉EPS发酵最优工艺,实验重复性好,是一个稳健、可控和高产的工艺技术方案,可为后续药理学研究和药物开发提供材料保障。Objective:To optimize the fermentation process of extracting exopolysaccharides(EPS)from Rhizopus nigricans in order to obtain the optimal fermentation process for EPS for the evidence support in following pharmacological research and drug development.Methods:Response surface methodology(RSM)was used to investigate the effects of pre-fermentation duration(the optimal time for temperature change),post-fermentation temperature and fermentation duration on the yield of EPS from Rhizopus nigricans to eventually determine the optimal batch fermentation process for EPS extraction.Results:RSM analysis showed that the factors influencing the response value was associated with the effectivity by fermentation duration>post-fermentation temperature>pre-fermentation duration.The factors producing significant impact on EPS production were associated with fermentation time(P<0.001)and post fermentation temperature(P=0.015).The optimal fermentation condition was 58.018 h for the process of pre-fermentation duration,26.984℃for post-fermentation temperature,and 143.193 h for fermentation duration.Under such a condition,the maximumly predicted yield of EPS was 0.891 g/L.Based on the equipment conditions and control capabilities,we had optimized the process as 58 h for pre-fermentation duration,27℃for post-fermentation temperature,and 143 h for fermentation duration.After verification of the process as described above,the yield of EPS was obtained by 0.889 g/L.Conclusion:RSM may be used to optimize EPS fermentation process for Rhizopus nigricans.The process optimized in this study can be a support in following pharmacological research and drug development because it is reproductive,controllable and high-yielding.
