摘 要:三磷酸腺苷(ATP)是一种重要的生物阴离子,其水平异常与各种疾病的发生发展密切相关。基于荧光团或荧光小分子的荧光分析方法因其操作简单、灵敏度高、选择性好等优点,已被广泛应用于ATP的检测中。但是目前许多荧光探针选择性较差,不能将ATP与其类似物(如其他核苷多磷酸盐等)区分开来,并且荧光剂容易聚集猝灭(ACQ)等问题,针对ATP的荧光传感技术仍然面临巨大挑战。基于此,本工作设计了一种金属有机框架结构/罗丹明衍生物复合探针(UiO-66/RhoB-N_(3)H_(5))。该探针在无ATP时是无荧光的。在ATP的诱导下,探针中的罗丹明-螺内酰胺结构会发生高效开环反应,于560 nm左右产生强烈的荧光发射。同时,金属有机框架结构(UiO-66)能够有效负载和分散罗丹明-螺内酰胺结构(RhoB-N_(3)H_(5)),大大提高了探针的稳定性和荧光响应强度。该探针对ATP表现出优良的灵敏度和选择性,线性范围为0.5~10 mM,检测限为0.1 mM,具备进一步应用于细胞内荧光成像的潜力。Adenosine triphosphate(ATP)is an essential biological anion,and abnormal ATP concentration is highly associated with the incidence and progression of various diseases.Fluorescent analysis methods based on fluorophores or small fluorescent molecules have been widely used in the detection of ATP because of their advantages of simple operation,high sensitivity and good selectivity.However,at present,the fluorescent sensing of ATP still faces great challenges because of the poor selectivity of many fluorescent probes,which cannot distinguish ATP from its analogues(such as other nucleoside polyphosphates),and the fluorescent agent is prone to the aggregation-caused quenching(ACQ).Based on this,a metal-organic framework structure/rhodamine derivative composite probe(UiO-66/RhoB-N_(3)H_(5))was designed in this work.The probe was non-fluorescent in the absence of ATP.Under the induction of ATP,the Rhodamine-spironolactamide structure in the probe would undergo an efficient ring-opening reaction,and produced strong fluorescence emission at around 560 nm.At the same time,the metal-organic framework structure(UiO-66)could be loaded and dispersed Rhodamine-spironolactamide structure(RhoB-N_(3)H_(5)),which greatly improved the stability and fluorescence response intensity of the probe.The probe showed excellent sensitivity and selectivity to ATP,with a linear range of 0.5~10 mM and a detection limit of 0.1 mM,which had the potential for further application in intracellular fluorescence imaging.
