迷迭香酸对乙醇诱导小鼠急性胃溃疡的保护作用探究

摘　　要：目的:探究迷迭香酸对乙醇诱导小鼠急性胃溃疡的保护作用及观察性别差异情况。方法:将30只昆明小鼠随机分为对照组(生理盐水注射组)、胃溃疡模型组、迷迭香酸低、高剂量组(腹腔注射迷迭香酸4 mg·kg^(-1)、20 mg·kg^(-1))、艾司奥美拉唑阳性对照组(腹腔注射3.03mg·kg^(-1)),每组6只,雌雄各半。除模型组外,各组小鼠分别预防性给药5 d,小鼠采用无水乙醇灌胃0.2 mL建立急性胃溃疡模型,观察小鼠行为学变化,1 h后麻醉处死小鼠,观察胃黏膜形态并计算溃疡指数;使用蛋白酶活性试剂盒测定胃蛋白酶活性等。结果:造模后,模型组小鼠出现了嗜睡,行动缓慢的行为学变化(达90%),而低、高剂量药物治疗组仅有少量出现了行为缓慢的现象(40%),提示迷迭香酸可能具有一定保护作用;无水乙醇模型组出现了明显的胃黏膜损伤,与对照组相比,胃溃疡指数明显升高(P<0.05),造模成功;低、高剂量迷迭香酸保护处理都能明显减轻乙醇对胃黏膜的损伤,减少胃黏膜出血以及溃疡的形成,与阳性药物作用相似,与模型组相比能显著降低胃溃疡指数(P<0.05),其中低剂量组作用更突出。胃蛋白酶活性各组有差别但无明显差异(P>0.05),迷迭香酸对雌、雄小鼠的保护作用无显著性差异(P>0.05)。结论:迷迭香酸对乙醇诱导的小鼠急性胃溃疡模型具有一定的保护作用,保护效果与艾司奥美拉唑保护效果相似,可减轻溃疡的形成。Objective:To explore the protective effect of rosmarinic acid on the acute gastric ulcer induced by absolute ethanol in mice and observe gender differences.Methods:A total of 30 Kunming mice were randomly divided into control group(normal saline injection group),gastric ulcer model group,rosmarinic acid low and high dose groups(intraperitoneal injection of rosmarinic acid 4 mg·kg^(-1),20 mg·kg^(-1)),and esomeprazole treatment positive control group(intraperitoneal injection of 3.03 mg·kg^(-1)),6 in each group,half male and half female.In addition to the model group,the mice in each group were administered prophylactically for 5 days,the mice were gavaged with 0.2 ml of absolute ethanol to establish an acute gastric ulcer model,the behavioral changes of the mice were observed,the mice were anesthetized and sacrificed after 1 hour,the gastric mucosal morphology was observed and the ulcer index was calculated,and the pepsin activity activity was determined by protease activity kit.Results:After modeling,the model group showed behavioral changes such as lethargy and slow movement(up to 90%),while only a small number of low-and high-dose drug treatment groups showed slow behavior(40%),suggesting that rosmarinic acid may have a certain protective effect.Compared with the control group,the gastric ulcer index was significantly increased(P<0.05),the modeling was successful;and the low-dose and high-dose rosmarinic acid groups could significantly reduce the damage of ethanol to the gastric mucosa,reduce the gastric mucosal bleeding and ulcer formation,which was similar to that of the positive control drug,the gastric ulcer index could be significantly reduced compared with the model group(P<0.05),and the effect of the low-dose group was more prominent.Pepsin activity was slightly different but not significantly among the groups(P>0.05).There was no significant difference in the protective effect of rosmarinic acid between female and male mice(P>0.05).Conclusion:Rosmarinic acid has a certain protective effect on ethanol
