摘 要:首次运用MISA工具从57.93 Gb的线口鳗基因组数据中搜索微卫星位点并分析其分布特征。结果显示,基因组中筛选得到总长度为14 044 174 bp的微卫星位点共计1 113 480个,这些微卫星位点随机分布在813 780条序列上,发生频率、出现频率和相对丰度分别为11.73%、16.05%和591.51个/Mb。在1~6个碱基重复的微卫星类型中,二碱基重复类型占比最大(558 274个,50.14%)、出现频率最高(8.05%)且相对丰度最大(296.57个/Mb)。在1 484种微卫星重复基元中,种类最多的为六碱基重复基元(603种),其次为五碱基重复(575种)、四碱基重复(230种)、三碱基重复(60种)、二碱基重复(12种)和单碱基重复(4种)。它们的优势碱基类别分别为A(178 791,47.83%)、CA(158 182,28.33%)、AAT(13 790,13.21%)、CACG(4 584,6.79%)、AATTG(166,3.71%)和CACACG(439,8.66%)。研究获得的线口鳗基因组中微卫星数量、种类和组成分布等信息,为今后开发利用线口鳗高质量的微卫星标记提供依据,也为进一步开展相关鱼类的种质遗传资源研究奠定基础。Inthis study,the MISA tool was used to search microsatellite sites from 57.93 Gb genomic sequencing data of Nemichthys scolopaceus for the first time.Microsatellite distribution characteristics were analyzed and the results showed that 1113480 loci with the length of 14044174 bp were obtained,which were randomly dispersed in 813780 scaffolds.The occurrence frequency,appearance frequency and relative abundance were 11.73%,16.05% and 591.51/Mb,respectively.Among the microsatellites with the repeat unit length of 1-6 bp,dinucleotide repeats accounted for the largest proportion(558274,50.14%),the highest appearance frequency(8.05%)and the greatest relative abundance(296.57/Mb).A total of 1484 microsatellite repeat motifs were detected,in which the hexanucleotide repeat motifs were the most abundant(603),and then followed by pentanucleotide(575),tetranucleotide(230),trinucleotide(60),dinucleotide(12)and mononucleotide(4)in turn.Their preponderant repeat motifs were A(178791,47.83%),CA(158182,28.33%),AAT(13790,13.21%),CACG(4584,6.79%),AATTG(166,3.71%)and CACACG(439,8.66%),respectively.Research data involving the number,distribution and abundance of different microsatellites in the genome of N.scolopaceus will provide basis for the development of high-quality microsatellite markers of this species in the future,and also lay the foundation for researches on germplasm resources of several related fishes.
