补中益气加味方对结直肠癌移植瘤模型细胞凋亡的影响

摘　　要：目的:探讨补中益气加味方对结直肠癌裸鼠移植瘤细胞凋亡及半胱氨酸天冬氨酸蛋白酶-3(Caspase-3)表达的影响。方法:将30只裸鼠构建SW480移植瘤模型,随机分为化疗组(腹腔注射氟尿嘧啶25 mg/kg)、模型组(0.9%氯化钠溶液)、中药低浓度组(26.72 g/kg)、中药中浓度组(53.43 g/kg)和中药高浓度组(106.86 g/kg),每组6只。其中,模型组和中药低、中、高浓度组裸鼠灌胃给药,1次/d,共14 d;化疗组隔天给药1次,共14 d。第15天处死裸鼠,统计比较各组裸鼠肿瘤体积及体质量,免疫组化法检测肿瘤细胞Caspase-3蛋白的表达情况,TUNEL法检测肿瘤细胞的凋亡。结果:5组裸鼠瘤体的体质量及体积进行比较,差异均无统计学意义(P>0.05);化疗组、中药中浓度组、中药高浓度组Caspase-3表达水平高于模型组,差异有统计学意义(P<0.05);化疗组与中药低浓度组、中药中浓度组、中药高浓度组Caspase-3表达进行比较,差异无统计学差异(P>0.05);中药低浓度组、中药中浓度组、中药高浓度组、化疗组细胞凋亡率均高于模型组,差异有统计学意义(P<0.05);中药中浓度组、中药高浓度组、化疗组细胞凋亡率显著高于中药低浓度组,差异有统计学意义(P<0.05);中药中浓度组和化疗组细胞凋亡率明显低于中药高浓度组,差异有统计学意义(P<0.05),中药中浓度组细胞凋亡率与化疗组比较,差异无统计学意义(P>0.05)。结论:补中益气加味方可提高结直肠癌裸鼠移植瘤模型肿瘤细胞凋亡及Caspase-3的表达。Objective:To explore the effect of Buzhong Yiqi Jiawei prescription on apoptosis and Caspase-3 expression of transplanted tumor cells in nude mice with colorectal cancer.Methods:Totally 30nude mice were constructed for the SW480 graft tumor model,and they were randomly divided into chemotherapy group(25 mg/kg intrabitoneal injection of fluorouracil),model group(0.9% sodium chloride solution),Chinese medicine low concentration group(26.72 g/kg),Chinese medicine medium concentration group(53.43 g/kg)and Chinese medicine high concentration group(106.86 g/kg),with 6 mice in each group.Among them,model group,Chinese medicine low concentration group,Chinese medicine medium concentration group and Chinese medicine high concentration group were given intragastric administration to nude mice,once a day for 14 days;The chemotherapy group was given the drug once every other day for 14 days.The nude mice were killed on the 15thday.Statistical comparisons were made for tumor volume and weight of each group.Caspase-3 protein expression was detected by immunohistochemistry method in tumor cells,and TUNEL assay was used to detect apoptosis of tumor cells.Results:There was no significant difference in tumor volume and weight among the 5groups(P>0.05);The chemotherapy group,Chinese medicine medium concentration group and Chinese medicine high concentration group showed higher expression of Caspase-3 than model group( P<0.05);There was no significant difference in expression of Caspase-3 among the chemotherapy group,and Chinese medicine low concentration group,Chinese medicine medium concentration group and Chinese medicine high concentration group(P>0.05).The Chinese medicine low concentration group,Chinese medicine medium concentration group,Chinese medicine high concentration group and chemotherapy group showed higher apoptosis rate than model group(P<0.05).The Chinese medicine medium concentration group,Chinese medicine high concentration group and chemotherapy group showed higher apoptosis rate than Chinese medicine low concen
