摘 要:目的:采用Box-Behnken响应面法优选中药复方降脂通脉方(JZTM)中多糖的提取工艺。方法:通过比较浸提法、回流法、超声法、超声辅助浸提法和超声辅助回流法5种方法,得出超声辅助回流法可提高多糖得率;之后,基于单因素实验优选JZTM多糖提工艺的影响因素及其范围,运用Box-Behnk响应面设计法构建三因素三水平实验方案,得出最佳工艺条件,再进行验证。结果:超声辅助回流法的多糖提取率最高,达到12.32 mg/g;进一步优化,得出最佳工艺条件为:液料比21.7∶1(mL/g),超声时间31.19 min,回流时间2.01 h,该条件下提取率达到27.14 mg/g。结论:该提取工艺简单可行,验证实验结果可靠,为JZTM的开发利用和工业化生产提供理论参考依据。Objective:To optimize the extraction process of polysaccharide from Jiangzhi Tongmai Formula(JZTM) by Box-Behnken response surface design.Methods:By comparing extraction method,reflux method,ultrasonic method,ultrasonic-assisted extraction method and ultrasonic-assisted reflux method,it was concluded that ultrasonic-assisted reflux method could improve polysaccharide yield.After that,the influencing factors and the range of JZTM polysaccharide extraction process were optimized based on single factor experiment,and the four-factor and three-level experiment scheme was constructed by Box-Behnk response surface design method,and the optimal process conditions were obtained,and then verified.Results:The extraction rate of polysaccharide by ultrasonic-assisted reflux method was the highest,reaching 12.32 mg/g.Further optimization,the optimum process conditions were as follows:liquid-solid ratio 21.7 ∶1(m L/g),ultrasonic time 31.19 min,reflux time 2.01 h.The extraction rate under this condition was 27.14 mg/g.Conclusion:The extraction process is simple and feasible,and the experimental results are reliable.It provides a reference for the development and utilization of Jiangzhitongmai formula and industrial production.
