摘  要:目的 探讨葫芦素B(Cucurbitacin B,CuB)对LPS诱导的C2C12肌管萎缩和C57BL/6小鼠骨骼肌萎缩的影响。方法 采用蛋白免疫印迹法检测蛋白表达水平,苏木精-伊红染色法观察细胞形态变化,通过测量小鼠握力以及腓肠肌和胫骨前肌质量评估骨骼肌萎缩程度。结果 蛋白免疫印迹试验表明,LPS能够诱导C2C12肌管萎缩(P<0.01),而经葫芦素B(0.3~3.0μmol/L)处理后可促进肌管分化相关蛋白(MyHC)及肌肉合成相关蛋白的表达,减少促炎因子(IL-1β、COX-2)释放(P<0.001)。苏木精-伊红染色试验结果从形态学角度证实了葫芦素B能有效改善LPS刺激下的肌管萎缩。此外,动物试验显示,LPS可导致C57BL/6小鼠握力以及腓肠肌与胫骨前肌质量显著下降,而经葫芦素B治疗后症状有所改善(P<0.05)。结论 葫芦素B在治疗骨骼肌萎缩方面具有潜在应用价值,为开发天然药物来源的先导化合物提供了理论依据,并为炎症诱导的骨骼肌萎缩的治疗开辟了新思路。Objective To explore the influence of Cucurbitacin B(CuB) on LPS-induced C2C12 myotube atro-phy and skeletal muscle atrophy in C57BL/6 mice.Methods Protein expression levels were determined by Western blot,cell morphological alterations were observed using H&E staining,and the degree of skeletal muscle atrophy was evaluated by measuring the grip strength of mice and the mass of gastrocnemius and tibialis anterior muscles.Results Western blot experiment indicated that LPS was capable of inducing the atrophy of C2C12 myotubes(P<0.01),while treatment with CuB(0.3~3.0 μmol/L) could promote the expression of myotube differentiation-related proteins(MyHC) and muscle synthesis-related proteins,and reduce the release of proinfla-mmatory factors(IL-1β,COX-2)(P<0.001).The results of H&E staining assay confirmed morphologically that CuB could effectively ameliorate the myotube atrophy stimulated by LPS.Additionally,the animal experiments revealed that LPS could lead to a significant decline in the grip strength of C57BL/6 mice and the mass of gastrocnemius and tibialis anterior muscles,and the symptoms were improved after treatment with CuB(P<0.05).Conclusion CuB holds potential application value in treatment of skeletal muscle atrophy.It offers a theoretical foundation for the development of lead compounds from natural medicinal sources and pioneers a novel thinking for the treatment of inflammatory skeletal muscle atrophy.
