摘 要:本试验以枯草芽孢杆菌(BS)为饲料添加剂,对脂多糖(LPS)应激下肉鸡肠道养分转运载体基因的表达情况进行研究。采用2×2双因素设计,因素1为枯草芽孢杆菌添加与否(0和200 g/t),因素2为LPS应激与否(0和500μg/kg BW)。选取体重相近、健康的1日龄爱拔益加肉鸡(公雏)192只,随机分为4组,每组设6个重复,每个重复8只鸡。对照组饲喂基础饲粮以及口服生理盐水;试验Ⅰ组饲喂基础饲粮以及口服LPS;试验Ⅱ组饲喂添加了枯草芽孢杆菌的基础饲粮以及口服生理盐水;试验Ⅲ组饲喂添加了枯草芽孢杆菌的基础饲粮以及口服LPS。试验分为应激前期(第1~14天)、LPS应激期(第15~21天)和应激后恢复期(第22~28天)3个阶段,共28 d。在使用第21天和第28天采集十二指肠、空肠和回肠样品,随后从样品中提取相关基因mRNA并进行反转录,然后通过荧光定量PCR进行基因表达的检测。结果显示:LPS应激使试验第21天时肉鸡整个肠道中B^(0)系统中性氨基酸转运载体(B^(0)AT1)、钠依赖性葡萄糖转运载体1(SGLT1)、脂肪酸转运载体4(FATP4)mRNA相对表达量显著降低(P<0.05);饲粮中添加枯草芽孢杆菌和LPS应激对肉鸡十二指肠中B^(0)AT1 mRNA相对表达量有显著的交互作用(P<0.05);饲粮中添加枯草芽孢杆菌显著提高了肉鸡空肠中SGLT1和FATP4 mRNA相对表达量(P<0.05)。饲粮中添加枯草芽孢杆菌显著提高了试验第28天时肉鸡回肠中B^(0)AT1 mRNA相对表达量(P<0.05)。综上可知,LPS应激会使肉鸡肠道中养分转运载体基因表达下调,饲粮中添加枯草芽孢杆菌可以上调肉鸡肠道中养分转运载体基因的表达,说明枯草芽孢杆菌能够缓解LPS应激对肉鸡肠道养分转运造成的负面影响,使肉鸡肠道养分转运载体基因正常表达。The aim of this experiment was to study the gene expression of intestinal nutrient transporter of broilers under lipopolysaccharide(LPS)stress using Bacillus subtilis as feed additive.A 2×2 two-factor design was adopted,factor 1 was whether Bacillus subtilis was added or not(0 and 200 g/t),and factor 2 was whether LPS was stressed or not(0 and 500μg/kg BW).A total of 192 healthy 1-day-old Arbor Acres broilers(male)with similar body weight were selected.They were randomly divided into 4 groups with 6 replicates per group and 8 chickens in per replicate.Broilers in control group were fed a basal diet and given normal saline through oral administration.Broilers in experimental groupⅠwere fed the basal diet and given LPS through oral administration.Broilers in experimental groupⅡwere fed the basal diet supplemented with Bacillus subtilis and given normal saline through oral administration.Broilers in experimental groupⅢwere fed the basal diet supplemented with Bacillus subtilis and given LPS through oral administration.The experiment was divided into three stages of pre-stress(days 1 to 14),LPS stress(days 15 to 21)and stress recovery(days 22 to 28),with a total of 28 days.Duodenum,jejunum and ileum samples were collected.Subsequently,the relevant mRNA was extracted from the samples and reverse-transcribed,and the expression of genes detected by fluorescent quantitative PCR.The results showed as follows:on day 21 of the experiment,LPS stress significantly reduced the mRNA relative expression levels of B^(0)system neutral amino acid transporter 1(B^(0)AT1),sodium-dependent glucose transporter 1(SGLT1)and fatty acid transporter 4(FATP4)in the whole intestine of broilers(P<0.05);there was a significant interaction between dietary supplementation of Bacillus subtilis and LPS stress on B^(0)AT1 mRNA relative expression level in duodenum of broilers(P<0.05);dietary supplementation of Bacillus subtilis significantly increased the mRNA relative expression levels of SGLT1 and FATP4 in jejunum of broilers(P<0.05).On da
