摘 要:本研究旨在探究棉籽糖(Raf)对黄曲霉毒素B_(1)(AFB_(1))诱导的小鼠肝脏氧化损伤的保护作用。将20只6~8周龄的C57BL/6雄性小鼠随机分为5组,分别为对照组、Raf组(给予50 mg/kg BW Raf)、AFB_(1)组(给予1.5 mg/kg BW AFB_(1))、低剂量Raf治疗组(给予25 mg/kg BW Raf和1.5 mg/kg BW AFB_(1))、高剂量Raf治疗组(给予50 mg/kg BW Raf和1.5 mg/kg BW AFB_(1)),每组4只。小鼠每隔1 d灌胃1次,共灌胃15次,为期30 d。灌胃期结束后进行生长性能、生化指标、相关蛋白表达检测以及组织切片观察。结果显示:1)Raf干预后,AFB_(1)诱导的小鼠摄食量减少、体重增长速度降低的趋势被抑制。2)Raf可以抑制AFB_(1)诱导的小鼠血清中谷草转氨酶(AST)和谷丙转氨酶(ALT)活性的极显著升高(P<0.01)。3)肝脏组织病理观察发现,与对照组相比,AFB_(1)组出现较多变性的细胞,并且排列不均匀,提示肝细胞受损,Raf干预后肝细胞受损情况得到改善。4)Raf可以抑制AFB_(1)诱导的小鼠肝脏中凋亡相关蛋白B细胞淋巴瘤-2(Bcl⁃2)蛋白表达水平的极显著降低(P<0.01)以及Bcl⁃2相关X蛋白(Bax)和半胱氨酸天冬氨酸蛋白酶-3(Caspase⁃3)蛋白表达水平的极显著升高(P<0.01)。5)Raf可以极显著降低AFB_(1)诱导小鼠肝脏中丙二醛(MDA)含量(P<0.01),极显著提高AFB_(1)诱导小鼠肝脏中超氧化物歧化酶(SOD)活性和还原型谷胱甘肽(GSH)含量(P<0.01)。6)Raf可以抑制AFB_(1)诱导的小鼠肝脏中氧化应激相关蛋白Kelch样ECH相关蛋白1(Keap1)蛋白表达水平的极显著升高(P<0.01)与血红素氧合酶-1(HO⁃1)蛋白表达水平的极显著降低(P<0.01)。综上所述,Raf对AFB_(1)诱导的小鼠体重增长速度降低、肝脏功能受损、肝脏氧化损伤具有改善作用。The aim of this study was to investigate the protective effect of raffinose(Raf)on the liver oxida⁃tive damage induced by aflatoxins B_(1)(AFB_(1))in mice.Twenty C57BL/6 male mice aged 6 to 8 weeks were randomly divided into 5 groups,they were control group,Raf group(50 mg/kg BW Raf was given),AFB_(1)group(1.5 mg/kg BW AFB_(1)was given),low dose RAF treatment group(25 mg/kg BW Raf and 1.5 mg/kg BW AFB_(1)were given),high dose Raf treatment group(50 mg/kg BW Raf and 1.5 mg/kg BW AFB_(1)were given),each group had 4 mice.Mice in each group were given a gavage per two days within 30 d.After gavage period,the growth performance,biochemical indexes,expression of related proteins were meas⁃ured and the tissue sections were observed.The results showed as follows:1)after Raf treatment,the feed in⁃take of AFB_(1)⁃induced mice was reduced,and the decreasing trend of weight gain rate was inhibited.2)Raf could inhibit the extremely significantly increase of serum aspartate aminotransferase(AST)and alanine amin⁃otransferase(ALT)activities induced by AFB_(1)(P<0.01).3)Pathological observation of liver tissue showed that,compared with the control group,there were more variable cells in AFB_(1)group with uneven arrange⁃ment,which indicated liver cell damage,and that was improved after Raf took effect.4)Raf could inhibit the extremely significantly decrease of liver apoptosis⁃related protein B⁃cell lymphoma⁃2(Bcl⁃2)protein expres⁃sion level(P<0.01)and the extremely significantly increase of Bcl⁃2⁃associated X protein(Bax)and cyste⁃ine⁃aspartic acid protease⁃3(Caspase⁃3)expression levels induced by AFB_(1)(P<0.01).5)RAF could signifi⁃cantly decrease the content of malondialdehyde(MDA)in liver of AFB_(1)⁃induced mice(P<0.01),and signif⁃icantly increase the activity of superoxide dismutase(SOD)and the content of reduced glutathione(GSH)in liver of AFB_(1)⁃induced mice(P<0.01).6)RAF could inhibit the extremely significantly increase of liver Kelch⁃like ECH⁃related protein 1(Keap1)protein e
