摘 要:目的:基于网络药理学探讨加减消瘰丸治疗肺结节病的作用机制。方法:通过中药系统药理数据库和分析平台(TCMSP)获取加减消瘰丸的主要化学成分及对应靶点,并使用UniProt数据库进行靶点与基因名称的转换;在OMIM、GeneCards数据库中检索肺结节病的疾病靶点并去重;通过Venny2.1.0在线工具进行映射取交集靶点;利用Cytoscape(v3.9.1)软件绘制“药物-化学成分-靶点”网络图,筛选发挥治疗作用的关键成分;利用STRING平台进行“加减消瘰丸-肺结节病”交集靶点互作分析,构建PPI网络,并使用Cytoscape(v3.9.1)对PPI网络处理,经分析得到加减消瘰丸作用于肺结节病的关键靶点;通过微生信平台对交集靶点进行GO和KEGG富集分析获知其潜在作用机制。结果:经筛选得到加减消瘰丸作用于肺结节病的有效成分42个和对应靶点225个,获得肺结节病的治疗靶点1530个,经靶点映射后得到加减消瘰丸治疗肺结节病的潜在靶点116个,关键成分有槲皮素、β-谷甾醇、山柰酚、柚皮素、豆甾醇、异鼠李素,关键靶点有TP53、AKT1、RELA、MAPK3、MAPK1、TNF、IL6等,关键通路包括TNF信号通路、IL-17信号通路、脂质与动脉粥样硬化通路等。结论:加减消瘰丸通过多成分、多靶点、多途径发挥治疗肺结节病的作用。This paper explores the action mechanism of modified Xiaoluo Pills in the treatment of pulmonary nodular disease based on network pharmacology.The main chemical constituents and the corresponding targets of modified Xiaoluo Pills were obtained from TCMSP database,and the targets and gene names were converted by using UniProt database.The targets of pulmonary nodular disease were retrieved and deduplicated in OMIM and GeneCards databases.The intersection of targets was mapped by using Venny 2.1.0 online tool.Cytoscape(v3.9.1)software was used to draw the drug-chemical component-target network diagram to screen the key components that play a therapeutic role.Using the STRING platform,the intersection target interactions of the drug and the disease was carried out,and the PPI network was constructed.Cytoscape(v3.9.1)was used to process the PPI network to obtain the key targets of modified Xiaoluo Pills in the treatment of pulmonary nodular disease.GO and KEGG enrichment analysis of the intersecting targets were carried out to find out the potential action mechanism.After screening,42 active ingredients and 225 corresponding targets of modified Xiaoluo Pills,and 1530 therapeutic targets of pulmonary nodular disease were obtained.After target mapping,16 potential targets of modified Xiaoluo Pills in the treatment of pulmonary nodular disease were obtained;the key ingredients included quercetin,β-sitosterol,kaempferol,naringenin,stigmasterol and isorhamnetin,the key targets included TP53,AKT1,RELA,MAPK3,MAPK1,TNF and IL6,and the key pathways included TNF signaling pathway,IL-17 signaling pathway,lipid and atherosclerosis pathway.In conclusion,modified Xiaoluo Pills exerts its therapeutic effect on pulmonary nodular disease through multi-components,multi-targets,and multi-pathways.
