摘 要:目的了解武汉市公园和农村生境中白纹伊蚊种群电压门控钠离子通道(VGSC)基因突变情况。方法2022年6—10月在武汉市2个公园和2个村湾分别采集白纹伊蚊幼虫和蛹,于实验室饲养至成蚊,抽提单只成蚊的基因组DNA,PCR扩增VGSC基因部分片段,测序后分析VGSC基因突变情况,并采用χ^(2)检验对种群间白纹伊蚊VGSC基因3个位点突变型等位基因的分布进行比较分析。结果共检测公园生境白纹伊蚊104只和农村生境白纹伊蚊43只。公园白纹伊蚊种群VGSC基因的1016、1532与1534位点均存在突变,且1016与1534位点及1532与1534位点均存在联合突变;而农村白纹伊蚊种群VGSC基因的1016与1534位点存在突变,1532位点未发现突变,且未发现联合突变现象。公园白纹伊蚊种群1534位点TCC/S和TGC/C突变型等位基因频率高于农村种群,差异具有统计学意义(P﹤0.001),但公园种群和农村种群间1016位点GGA/G突变型等位基因与1532位点ACC/T突变型等位基因的突变频率差异均无统计学意义(P=0.160,P=0.263)。结论公园和农村白纹伊蚊种群VGSC基因突变情况有差异,建议开展白纹伊蚊VGSC基因突变监测,以进一步了解武汉市白纹伊蚊拟除虫菊酯类杀虫剂抗药性迅速发展的作用机制。Objective To investigate the voltage-gated sodium channel(VGSC)gene mutations of Aedes albopictus in park and rural habitat in Wuhan City.Methods Larvae and pupae of Ae.albopictus were collected from 2 parks and 2 villages in Wuhan City during July to October 2022 and then raised to adult mosquitoes in the laboratory.Genomic DNA was extracted from individual mosquito and the partial sequence of VGSC gene fragments were amplified and sequenced to detect the mutation.The distribution of mutant alleles at 3 loci of VGSC gene were compared between different Ae.albopictus populaitons by Chi-square test.Results A total of 104 Ae.albopictus from park habitat and 43 Ae.albopictus from rural habitat were analyzed.Mutations were detected at the 1016,1532 and 1534 loci of VGSC gene in park population,where joint mutant alleles at two of the three loci(i.e.1016+1532,1532+1534 loci)were found.Anyway,mutations were detected at the 1016 and 1534 loci but not at 1532 loci of VGSC gene in rural population,where no joint mutation was observed.The rate of mutant allele TCC/S and TGC/C at 1534 loci was significant higher in the park population than in rural population,with P<0.001,while no significant difference was found in the rate of mutant allele GGA/G at 1016 loci or mutant allele ACC/T at 1532 loci between the two Ae.albopictus field populations,with P=0.160 and P=0.263,respectively.Conclusion The rate of VGSC gene mutation was various between the park Ae.albopictus population and rural Ae.albopictus population.It is suggested to monitor the VGSC gene mutation to better understand the mechanism of rapidly developed pyrethroid insecticides resistance in Ae.albopictus.
