摘  要:类甜蛋白(Thaumatin-like proteins,TLPs)是一类与甜蛋白(Thaumatin)高度同源且具有抗真菌活性的蛋白家族,广泛分布于多种植物、动物及微生物。为了解全基因组水平上花生类甜蛋白家族的基本生物学特征,本研究通过生物信息学手段,鉴定了花生类甜蛋白家族成员,并对其基因结构、染色体定位、进化、基因表达模式、蛋白结构及蛋白质互作网络进行了分析。结果显示,花生中鉴定到62个类甜蛋白家族成员,分布于花生全部的20条染色体上,同一染色体上存在串联重复的TLP基因簇。蛋白序列进化分析表明,花生62个类甜蛋白分为10个聚类,与豆类亲缘关系较近,进化上高度保守。基因表达模式分析显示位于2号染色体的花生类甜蛋白在胚胎组织中有很高的表达量,说明其在胚胎发育早期起到一定作用。同时,位于15号染色体的花生类甜蛋白在根及根尖组织中的表达量明显高于其他组织,结合类甜蛋白在豆科植物结瘤中的作用,说明该组基因簇可能在花生结瘤过程中发挥相应作用。蛋白结构分析表明,花生AhTLP1含有3个保守的类甜蛋白结构域,同时在该蛋白N端和C端各含有一个跨膜结构域。蛋白质互作分析表明花生AhTLP1蛋白与10个不同功能的蛋白质相互作用,其中AhTLP1与PP2C(Protein phosphatase type 2C)的相互作用可能使其参与到ABA信号转导的途径中,从而提高该蛋白在植物抵御非生物胁迫中的作用。该研究为花生类甜蛋白基因家族的功能分析和遗传育种应用提供了基础。Thaumatin-like proteins(TLPs),a group of highly homologous proteins to thaumatin,have antifungal activity and are widely distributed in a variety of plants,animals,and microorganisms.To understand the basic biological characteristics of the peanut thaumatin-like protein family at the whole genome level,the members of the peanut thaumatin-like protein family were identified and their gene structure,chromosome localization,gene expression pattern,and protein interaction network were analyzed.The results showed that 62 members of the thaumatin-like protein family were identified and distributed on all 20 chromosomes of peanut with tandem and repeated TLP gene clusters.The analysis of protein sequence evolution showed that 62 TLPs of peanut were divided into ten clusters,which were closely related to legumes and highly conserved in evolution.Gene expression pattern analysis showed that peanut thaumatin-like protein located on chromosome 2 had high expression level in embryonic tissue,indicating that it playes a certain role in early embryonic development.At the same time,the expression of peanut thaumatin-like protein located on chromosome 15 was significantly higher in root and root tip tissues than other tissues,indicating that this gene cluster played a specific role in the process of peanut nodulation.Protein structure analysis showed that peanut AhTLP1 contained three conserved sweet protein-like domains,and there is a transmembrane domain at both of the N-terminal and the C-terminal of the protein.Protein interaction analysis showed that peanut AhTLP1 protein interacts with 10 proteins with different functions,and the interaction between AhTLP1 and protein phosphatase type 2C(PP2C)may involve AhTLP1 in ABA signal transduction pathway for its abiotic stress resistance improvement.In summary,this study provides a basis for the functional analysis and genetic breeding application of peanut thaumatin-like protein gene family.
