摘 要:丰富糜子种质资源,可以为糜子育种和功能基因组学研究提供新材料。采用EMS诱变技术,构建糜子突变体库。以河北高产、优质糜子品种冀黍3号为试验材料,采用二因素(EMS溶液浓度、浸种时间)三水平(EMS溶液浓度0.6%、0.8%和1.0%,浸种时间8、10、12 h)试验设计,以不进行EMS诱变为对照,对糜子EMS诱变条件进行优化;并在此条件下进行糜子突变体库构建,在M_(2)代进行表型性状鉴定,在M4代进行表型性状验证鉴定。结果表明:糜子EMS诱变的最优条件为EMS溶液浓度0.8%、浸种10 h;经M4代表型性状验证,获得稳定遗传的13个表型突变性状,共计139个突变材料,突变频率为3.53%。Enriching the germplasm resources of proso millet can provide new materials for breeding and functional genomics research.Using EMS mutagenesis technology,a mutant library of proso millet was constructed.Taking high-yield and high-quality proso millet variety Jishu 3 in Hebei as the experimental material,a two-factor(EMS solution concentration and soaking time)three-level(EMS solution concentrations of 0.6%,0.8%,and 1.0%,and soaking time of 8,10,and 12 h)experimental design was adopted to optimize the EMS induction conditions of proso millet,taking without EMS induction as the control.Under these conditions,a mutant library of foxtail millet was constructed,and the phenotypic traits were identified in the M_(2)generation and validated in the M4 generation.The results showed that the optimal conditions for EMS mutagenesis of foxtail millet were EMS solution concentration of 0.8%and soaking for 10 h.After verification of M4 representative traits,13 mutant phenotypic traits and 139 stable genetic mutant materials were obtained,with a mutation frequency of 3.53%.
