摘  要:为了研究植物隐花色素基因(CRY)在高粱生长发育、胁迫响应中所起的作用,首先,利用全基因组BLAST对高粱基因组进行分析挖掘潜在CRY同源基因,进一步利用在线网站ExPASy和TBtools软件对其理化性质、基因结构、进化关系、基因共线性、启动子顺式调控元件进行分析;同时,采用高粱公共转录组数据库及qRT-PCR对其组织表达模式和在不同环境胁迫下的响应模式进行研究。结果表明:通过基因家族鉴定,得到9个SbCRYs家族成员,分布在5条染色体上,命名为SbCRY1—SbCRY9。该基因家族氨基酸长度为447~710个,蛋白质分子质量为48.29~110.13 kD,等电点为5.53~9.44。进化分析结果显示,该家族与拟南芥、水稻、谷子、玉米的CRY成员一起可被分为6个亚家族。SbCRYs成员之间仅存在1对同源基因对,而与水稻、谷子、玉米之间分别存在8、9、11对同源基因对。通过对SbCRYs基因启动子区进行分析,发现存在着激素和环境胁迫响应元件,且这些基因存在明显的组织表达特异性。高粱CRY家族成员能够不同程度地受到高温胁迫、盐胁迫、干旱胁迫和蚜虫侵害的诱导表达,其中SbCRY2、SbCRY3和SbCRY4参与了盐和干旱胁迫的响应,SbCRY2、SbCRY3、SbCRY4、SbCRY6、SbCRY7和SbCRY9参与了高粱蚜虫胁迫的响应。这些结果揭示了高粱SbCRYs基因在不同组织中可能的功能以及对非生物胁迫的响应。In order to investigate the role of the plant cryptochrome(CRY)gene in the growth,development and stress response of sorghum,first,the sorghum genome was analyzed using whole genome BLAST to identify potential CRY homologous genes.Further,the physical and chemical properties,gene structure,evolutionary relationships,gene collinearity,and promoter cis regulatory elements were analyzed using online websites ExPASy and TBtools software.At the same time,the sorghum public transcriptome database and qRT PCR were used to study its tissue expression patterns and response patterns under different environmental stresses.The results showed that through gene family identification,nine SbCRYs family members were obtained,distributed on 5 chromosomes,and named SbCRY1-SbCRY9.The amino acid length of this gene family is 447-710,the protein molecular weight is 48.29-110.13 kD,and the isoelectric point is 5.53-9.44.The evolutionary analysis results showed that this family,along with the CRY members of Arabidopsis,rice,foxtail millet,and maize,can be divided into six subfamilies.There is only one pair of homologous gene pairs among SbCRYs family members,while there are 8,9,and 11 pairs of homologous gene pairs with rice,foxtail millet,and maize,respectively.By analyzing the promoter region of the SbCRYs gene,it was found that there are hormone and environmental stress response elements present,and these genes exhibit significant tissue expression specificity.Members of the SbCRYs family members can be induced to express under varying degrees of high temperature stress,salt stress,drought stress,and aphid invasion.Among them,SbCRY2,SbCRY3,and SbCRY4 are involved in the response to salt and drought stress,while SbCRY2,SbCRY3,SbCRY4,SbCRY6,SbCRY7,and SbCRY9 are involved in the response to sorghum aphid stress.These results reveal the possible functions of sorghum SbCRYs genes in different tissues and their response to abiotic stress.
